DNA strand specificity for UV-induced mutations in mammalian cells.

Abstract: The influence of DNA repair on the molecular nature of mutations induced by UV light (254 nm) was investigated in UV-induced hprt mutants from UV-sensitive Chinese hamster cells (V-Hi) and the parental line (V79). The nature of point mutations in hprt exon sequences was determined for 19 hprt mutants of V79 and for 17 hprt mutants of V-Hi cells by sequence analysis of in vitro-amplified hprt cDNA. The mutation spectrum in V79 cells consisted of single-and tandem double-base pair changes, while in V-Hi cells th… Show more

“…Another example of higher mutagenesis in the TS was observed previously in mfd-deficient E. coli lacking the TCR process (Oller et al 1992). These results are in contrast with those of the TSs of RNAP II genes that are preferentially repaired and, consequently, show a lower mutagenesis as compared with the NTSs (Vrieling et al 1989;McGregor et al 1991;Sage et al 1993). In conclusion, these data establish a direct link between NER and mutagenesis, and indicate that RNAP III transcription is not coupled to NER and that, in opposition to RNAP II genes, it is the NTSs of RNAP III genes that are preferentially repaired.…”

“…But evidence does exist linking mismatch repair activities with TCR (4,17,22); thus, the factors regulating the process of gene repair remain to be fully defined. For example, an elevated number of mutations have been observed to form initially on the nontranscribed strand of the human HPRT gene, a gene transcribed by RNA polymerase II (34,35). Thus, enzymatic processing events during transcription can actually be focused toward the nontranscribed strand.…”

“…In general, the e ects of transcription on repair are more pronounced at low levels of DNA damage (Vrieling et al, 1989(Vrieling et al, , 1991. Since much higher levels of DNA damage are typically employed for repair assays than for mutation assays, this may be the reason that the strand bias of mutation is much more pronounced than the strand bias of repair.…”
