DNA Methyltransferase 1 Knock Down Induces Gene Expression by a Mechanism Independent of DNA Methylation and Histone Deacetylation

Abstract: DNA methyltransferase 1 (DNMT1) catalyzes the postreplication methylation of DNA and is responsible for maintaining the DNA methylation pattern during cell division. A long list of data supports a role for DNMT1 in cellular transformation and inhibitors of DNMT1 were shown to have antitumorigenic effects. It was long believed that DNMT1 promoted tumorigenesis by maintaining the hypermethylated and silenced state of tumor suppressor genes. We have previously shown that DNMT1 knock down by either antisense oligo… Show more

“…These findings suggest that inhibition of DNA methylation may actually reduce the expression of specific genes, which may explain reduced H3 acetylation that Glen Schafe's group observed after DNMT inhibition (Monsey et al, 2011). In addition, DNA methylation-independent effects of DNMTs have been noted (Milutinovic et al, 2004), as have direct interactions between DNMTs and HDACs (Robertson, 2002). Thus, further studies of gene-specific changes in DNA methylation are required to strengthen the conclusion that memory impairment caused by DNMT inhibitors can be attributed to changes in DNA methylation.…”

“…DNMT1 interacts directly with HDAC 1 and 2 as well as the histone methyltransferase SUV39H1 (Vaute et al, 2002). DNMT1 can also activate Sp1 response elements without increasing the protein levels of of Sp1 or Sp3 or even the occupancy of their elements by these proteins (Milutinovic et al, 2004).…”

“…16 In general, DNMT1 is responsible for maintaining DNA methylation patterns during cell division, 13 while DNMT3b and DNMT3a are associated with de novo methylation. 17 In addition to DNA methylation, histone modification also helps organize nuclear architecture for the regulation of gene expression and serves as a dynamic regulator of gene activities. 18 Unlike DNA methylation, covalent histone modifications are more unstable and diverse; these include post-translational modifications as acetylation, phosphorylation, methylation, sumoylation and ubiquitylation.…”
