DNA Methylation Detection and Inhibitor Screening Based on the Discrimination of the Aggregation of Long and Short DNA on a Negatively Charged Indium Tin Oxide Microelectrode

Abstract: On the basis of the different diffusivity and existence of electrostatic repulsion between long and short DNA on the negatively changed indium tin oxide (ITO) microelectrode, a simple but sensitive immobilization free solution-phase electrochemical method for DNA methylation detection and inhibitor screening has been developed. Electroactive substance (methylene blue) tagged at the penultimate base T close to the 3'-terminal first, in the absence of DNA methylation, methylene blue-labeled electroactive fragmen… Show more

“…Then, the methylated dsDNA is cleaved by DpnI. According to the T m value calculation equation (T m = 4 • C × G/C pair + 2 • C × A/T pair) [25], the remained hybrid only had a T m of 22 • C and will be detached into a single-stranded DNA (ssDNA) at the reaction temperature (37 • C). Hence, the Gquadruplex DNAzymes can be formed between two short ssDNA when hemin is introduced, so an obvious DPV signal of benzoquinone originated from the oxidation of hydroquinone by H 2 O 2 can be detected.…”

“…Among them, electrochemical method, as a useful analytical technology, has been extensively employed because of its outstanding merits of inexpensive, fast detection, convenient operation procedures, high sensitivity, and compatibility with micro-manufacturing techniques 17, 18 . In particularly, recently developed homogeneous electrochemical methods open a promising way for the design of immobilization-free electrochemical biosensors 1921 . In these biosensors, the recognition of analyte and subsequent reaction are performed in homogeneous solution instead of the interface between the solution phase and electrode surface.…”

“…Cai reported several electrochemical method using signal amplification methods (Li et al, 2012;Liu et al, 2011). Plenty of methods based on bio enzyme are also reported (Muren and Barton, 2013;Mao et al, 2013;Ronen et al, 2014;Wei et al, 2014). Another important type of methods are based on methyl CpG-binding domain (Yin et al, 2013;Zhou et al, 2014b) or specific immuno-recognition of methyl CpG (Kurita and Niwa, 2012;Wang et al, 2012;Yin et al, 2013).…”
