DNA‐Mediated Assembly of Gold Nanoparticles and Applications in Bioanalysis

Abstract: DNA-mediated gold nanoparticles (AuNPs) assembly has drawn great attentiond ue to the controllable parameters and properties. In general,D NA sequences working as linkerso rt emplates precisely controlt he assembly of AuNPs throught he Watson-Crick base pairing. Diverse assembly structures show great potential in bioanalysis due to the uniqueo ptical characteristics, which can be tuned by the shape, size, composition, particles interactive statusa nd the dielectric constant of the surrounding mediumo f AuNPs.T… Show more

“…[25,26,[206][207][208] To prepare small NP dimers or trimers, multidentate ligands (e.g., dithiolates) are used to link together constituent particles. [74,209] To precisely assemble more complex 2D or 3D oligomers such as tetramers, pentamers, hexamers, heptamers, etc., the directional interactions between NPs introduced by polymer [143,210] or DNA ligands, [211][212][213] typically thiolated, [212] may be required. Designed ligands can be synthesized and introduced to the NP surface through ligand exchange, described in Section 2.…”

“… 14 , 15 , 16 , 17 Among them, AuNPs have become one of the most widely used nanomaterials in the construction of DNA nanomachines. 18 AuNPs, usually in the size of 1–100 nm, could display unique optical properties by adjusting their size and shape, rendering them a typical nanomaterial for surface plasmon resonance (SPR) and surface-enhanced Raman scattering (SERS) analysis. 19 , 20 The aggregation state of AuNPs can indicate the color change of the solution.…”

“…These unique properties enable nanomaterials to be used as sensing probes, contrast agents, therapeutic agents, and drug delivery systems that provide a new modality to solve the current clinical tumor treatment problems. [2][3][4][5][6] Nanomaterials are widely used in fundamental research concerning cancer diagnosis and therapy to make nanotechnology feasible for clinical application. It is highly meaningful to integrate different imaging and treatment methods into one system and construct a multifunctional nanostructure system for imaging, drug delivery, and therapy.…”
