DNA markers to disentangle complexes of cryptic taxa in mealybugs (Hemiptera: Pseudococcidae)

Abstract: Mealybugs (Hemiptera: Pseudococcidae) are major pests of a wide range of crops and ornamental plants worldwide. Their high degree of morphological similarity makes them difficult to identify and limits their study and management. We aimed to identify a set of markers for the

“…The sequence comparison, after alignment (Fig 4 and 5), reveals the extent of divergence corresponding to interspecific variations in previous studies (Malausa et al 2011;Park et al 2011). At COI, the divergence was >7% between D. lavandulae and D. multivorus and >9% between D. lavandulae and the Trionymus species.…”

“…The third one, hereafter referred to as COI, is located in the mitochondrial region of Cytochrome Oxidase Subunit I. For COI, we amplified three overlapping fragments previously used in several molecular studies on Pseudococcidae (Malausa et al, 2011;Park et al, 2011;Abd-Rabou et al, 2012), covering in total ~1050 bp. The sequences of the primers used to amplify each region are provided in Table 1. PCR were performed using the Qiagen Multiplex PCR kit (QIAGEN, Hilden, Germany), in a reaction volume of 25 µL.…”

“…Morphological identification of the species was made by J.-F. Germain (ANSES, Laboratoire National de la Protection des Végétaux, Montpellier, France). The species identification was ascertained by molecular characterization using a set of markers able to efficiently distinguish taxa within Pseudococcidae (22). Insects were maintained in darkness at 23°C (±2°C) in plastic jars whose lids had been perforated and covered with a 28 µm mesh tissue to enable air circulation.…”
