DNA macroarray study of skin aging‐related genes expression modulation by antioxidant plant extracts on a replicative senescence model of human dermal fibroblasts

Abstract: The formation of reactive oxygen species (ROS) is a widely accepted pivotal mechanism leading to skin aging. It increases with age, while the endogenous defense mechanisms that counter them decrease. This imbalance, called oxidative stress, leads to the progressive damage of cellular structures and results in accelerated aging. Antioxidant compounds can provide protection from endogenous and exogenous oxidative stress by scavenging free radicals. The main phenolic compounds of oak wood, mate leaf and benjoin r… Show more

“…Additionally, the expression of the E2F4 was higher after the benjoin resin treatment, in the comparison to oak wood and mate leaf phenolics action. Significant up-regulation was also noted for analyzed extracts in the antioxidant catalase gene [ 122 ].…”

“…Cutaneous aging is a complex phenomenon involving two simultaneously occurring processes: intrinsic aging and extrinsic aging. Intrinsic aging, known as chronological or natural aging, is genetically determined, whereas extrinsic aging is caused by environmental factors, such as chronic sun exposure (the total sunlight spectrum contains 45% ultraviolet light), and is known as photoaging [ 53 ]. The main symptoms of dermal aging are represented by the gradual process of wrinkle development, combined with skin sagging and drooping.…”

“…Both types of cutaneous aging reduce the proliferative capacity of fibroblasts, keratinocytes, and melanocytes [ 53 ]. The morphological manifestations are the result of a decreased extracellular matrix (ECM) synthesis in the dermis due to the increased expression of matrix-degrading enzymes and matrix metalloproteinases (MMPs), which are mainly secreted by epidermal keratinocytes and dermal fibroblasts [ 55 ].…”

“…On the 39 other hand, synthetic phenolic antioxidants such as butylated hydroxyanisole (BHA), 40 butylated hydroxytoluene (BHT), tert-butylhydroquinone (TBHQ), and propyl gallate 41 (PG) (Figure 1) are well known as food preservatives or antioxidants and have been M A N U S C R I P T A C C E P T E D ACCEPTED MANUSCRIPT 5 broadly used for extending the shelf-life of lipid-rich food products (Li,Ji,Sun,Yang,& 43 Chu, 2009). 44 Traditionally, the antioxidant activity of a compound is assessed by measuring 45 scavenging activity against DPPH or other free radicals activity and reducing power 46 assays (Dudonné, Vitrac, Coutière, Woillez, & Mérillon, 2009). However, the results of 47 those chemical-based spectrophotometric assays may not be closely linked with the real 48 antioxidant activity of inhibiting the lipid oxidation occurring in food and human body.…”
