DNA/lysozyme binding propensity and nuclease properties of benzimidazole/2,2′-bipyridine based binuclear ternary transition metal complexes

Abstract: The binuclear ternary transition metal complexes have the ability to bind to lysozyme and DNA as well as DNA cleavage. Complexes exhibited cytotoxicity to only the cancerous cells (HepG-2) and is safe to the non-malignant HEK-293.

“…Additionally, DNA intercalation was studied by classic competitive binding assays with ethidium bromide . Most coordination compounds can intercalate DNA with K app ≈ 10 5 M, and the values obtained correlate with those obtained for coordination compounds of copper with anticancer activity . Expectedly, organic ligands do not intercalate DNA, except for cytotoxic ligand 16 (Table S25 and Figures S35–S37).…”

“…42 Most coordination compounds can intercalate DNA with K app ≈ 10 5 M, and the values obtained correlate with those obtained for coordination compounds of copper with anticancer activity. 43 Expectedly, organic ligands do not intercalate DNA, except for cytotoxic ligand 16 (Table S25 and Figures S35−S37). However, a lack of correlation between the ability to intercalate DNA and cytotoxicity makes the DNA intercalation ability noncritical for the cytotoxicity of studied ligands and coordination compounds.…”

“…In 1952, the first reported studies regarding the biological activity of cobalt complexes examined the toxicity of the optical isomers of [Co(en) 3 ](NO 3 ) 3 (en = ethylenediamine) [ 3 ]. Since then, many cobalt complexes of biological interest have been reported with the majority of them showing antiproliferative [ [4] , [5] , [6] , [7] ], antimicrobial [ [8] , [9] , [10] ], antifungal [ 11 , 12 ], antiviral [ 13 , 14 ], and antioxidant [ [15] , [16] , [17] ] activity.…”

“…Form I, one of the three bands formed as a result of agarose gel electrophoresis, moves faster than Form II and Form III. This is due to the large load density and low volume (Mansour and Ragab 2019 ). According to Sohrabijam et al ( 2017 ), they investigated the effect of chitosan coated superparamagnetic nanoparticles on DNA molecules.…”
