In the present study, we attempt to confirm the effect of the ethyl acetate fraction from Aruncus dioicus var. kamtschaticus (EFAD) against neurodegeneration with various experiments using trimethyltin (TMT). First, learning and memory deterioration caused by TMT was examined with several behavioral tests. EFAD had an improvement effect on TMT‐induced cognitive dysfunction. Second, to assess the oxidative stress caused by TMT, a series of biochemical indicators of the cholinergic and antioxidant system were measured. EFAD showed superb effects on the recovery of the cholinergic system and reduction of oxidative stress. Third, to identify the mitochondrial apoptosis caused by TMT, mitochondrial activity and mitochondrial apoptotic signaling molecules were analyzed. EFAD improved mitochondrial activity and suppressed mitochondrial apoptosis. Finally, dicaffeoylglucose isomers were identified as main compounds of EFAD with ultra‐performance liquid‐quadrupole‐time‐of flight mass spectrometry (UPLC‐QTOF/MS2). Consequently, EFAD showed a protective effect against mitochondrial apoptosis‐induced neurodegeneration in TMT‐injected mice based on antioxidant ability. Practical applications Recently, neurodegenerative disease patients are increasing, and in particular, Alzheimer’s disease (AD) is a representative neurodegeneration, which characterized by mitochondrial damage, neurofibrillary tangles, and neuronal cell death. This study confirmed that the ethyl acetate fraction from A. dioicus var. kamtschaticus ameliorates the impaired cognitive ability by TMT toxicity, which shares the symptoms of AD. Therefore, this study can provide the potential of cognitive functional food material of A. dioicus var. kamtschaticus.

Aruncus dioicus var. kamtschaticus is an economically important herb in the cold temperate regions of East Asia, and displays highly variable morphological features. Completed chloroplast genome of A. dioicus var. kamtschaticus isolated in Korea is 157,859 bp long with four subregions: 85,972 bp of large single copy and 19,185 bp of small single-copy regions separated by 26,351 bp of inverted repeat regions. The genome includes 131 genes (86 protein-coding genes, eight rRNA s, and 37 tRNA s). Phylogenetic analyses show that our chloroplast genome was clustered with two partial chloroplast genomes of A. dioicus .

This study was performed to estimate the possibility of using an ethyl acetate fraction from Aruncus dioicus var. kamtschaticus (EFAD) on metabolic syndrome that is induced by a high-fat diet (HFD). It was demonstrated that EFAD suppresses lipid accumulation and improves insulin resistance (IR) caused by Tumor necrosis factor alpha (TNF-α) in in-vitro experiments using the 3T3-L1 cell. In in-vivo tests, C57BL/6 mice were fed EFAD at 20 and 40 mg/kg body weight (BW) for four weeks after the mice were fed HFD for 15 weeks to induce obesity. EFAD significantly suppressed the elevation of BW and improved impaired glucose tolerance in obese mice. Additionally, this study showed that EFAD has an ameliorating effect on obesity-induced cognitive disorder with behavioral tests. The effect of EFAD on peripheral-IR improvement was confirmed by serum analysis and western blotting in peripheral tissues. Additionally, EFAD showed an ameliorating effect on HFD-induced oxidative stress, impaired cholinergic system and mitochondrial dysfunction, which are interrelated symptoms of neurodegeneration, such as Alzheimer’s disease and central nervous system (CNS)-IR in brain tissue. Furthermore, we confirmed that EFAD improves CNS-IR by confirming the IR-related factors in brain tissue. Consequently, this study suggests the possibility of using EFAD for the prevention of neurodegeneration by improving metabolic syndrome that is caused by HFD.
