DNA binding and photocleavage properties of two mixed-ligand oxovanadium complexes

Abstract: An oxovanadium complex [VO(satsc)(bipy)](1) (satsc = salicylaldehyde thiosemicarbazone, bipy = 2,2 0 -bipyridine) and its dibromo derivative [VO(3,5-dibrsatsc)(bipy)] (2) (3,5-dibrsatsc = 3,5-dibromosalicylaldehyde thiosemicarbazone) have been synthesized and characterized by elemental analysis, IR, ES-MS and 1 H NMR. The interaction of these two complexes with calfthymus DNA (CT-DNA) was studied using UV/Vis, fluorescence spectroscopic titration, viscosity measurements and thermal denaturation. The results su… Show more

“…The complexes show an intense band at ca. 265 nm assignable to π - π * transitions of aromatic rings of phenanthroline [1820, 28]. A medium band is observed near 400 nm, which is attributed to a ligand-to-metal charge-transfer transition (LMCT) as a charge transfer from a p-orbital on the lone-pair of ligands oxygen atoms to the empty d-orbital of the vanadium atom [27, 29].…”

“…On the other hand, vanadyl(IV) complexes incorporating thiosemicarbazones have been studied extensively for their insulin-like effects which result in the inhibition of glycerol release and enhancement of glucose uptake by rat adipocytes and have been used in the treatment of tuberculosis [1620]. In view of inquisitive response of oxovanadium(IV) in biology, we have reported that four oxidovanadium(IV) complexes present highly cytotoxic activities against Myeloma cell (Ag8.653) and Gliomas cell (U251) lines [21].…”

“…Yet, the detailed mechanisms by which such complexes inhibit the growth of cancer cells have not been clearly elucidated. Our previous results also indicated that upon increasing the appending aromatic moiety, the biological activities of oxidovanadium complexes tend to be better (26,27,31). It seems to us that the introduction of some novel ligands may be a useful tool in moderating the antitumor effects of vanadium complexes (32).…”

“…The CT-DNA in buffer gave a ratio of UV absorbance at 260 and 280 nm of 1.8-1.9, indicated the DNA free of protein [43][44][45] [complex] produced a slope of 1/(εb -εf), and a Y intercept equal to 1/Kb(εb -εf); Kb is the ratio of slope to the Y intercept [46].…”
