DNA-Based Immunization against Lyssaviruses

Abstract: Rabies is a fatal encephalomyelitis. Most cases occur in developing countries and are transmitted by dogs. Because of their high cost, cell culture vaccines have not totally replaced the unsafe brain-derived vaccines which are still used in many developing countries. Moreover, there will be a need for vaccines against rabies-related viruses against which classical vaccines are not always effective. DNA vaccines would, therefore, be a valuable alternative for the production of cheaper rabies vaccines against a … Show more

“…This response is related to the presence of antibodies. IL-4 detection has previously been reported by Perrin et al (25) after administration of a naked DNA rabies vaccine in dogs. Moreover, the production of IFN and high antibody titres in sheep vaccinated with a recombinant adenovirus and with an edible vaccine that express the GpVR have been demonstrated (2) .…”

“…Esta respuesta está relacionada con la presencia de anticuerpos. La detección de IL-4 fue previamente informada por Perrin et al (25) después de la administración de una vacuna de ADN desnudo contra la rabia en perros. Por otra parte se ha demostrado la producción de IFN- y altos títulos de anticuerpos en ovejas vacunadas con un adenovirus recombinante y con una vacuna comestible que expresa GpVR (2) .…”

“…Antigen expression is increased when an untranslated region, with its splicing signals, follows the promoter. 26 The sequence encoding the antigen is 3 -flanked by the transcript termination/polyadenylation (polyA) signal of the SV40 or of the bovine growth hormone (BGH). The plasmid pcDNA3 and its derivatives, which contain the HCMVie promoter and the BGH polyA, have often been used as vectors for DNA vaccines (Fig.…”
