DNA analysis of HLA–DR and DQ genes in american blacks with systemic lupus erythematosus

Abstract: We studied DNA polymorphisms of HLA-DR and DQ alleles in 63 American black patients with systemic lupus erythematosus (SLE). We found no HLA-DR beta, DQ alpha, or DQ beta restriction fragment length polymorphism (RFLP) or RFLP-determined DR or DQ specificity associated with SLE in either the patients or in 57 control subjects. DRw52b was positively associated with renal involvement and negatively associated with anti-nuclear RNP antibodies. Antibodies to Ro (SS-A) and La (SS-B) were associated with DR3(DRw17),… Show more

“…We did, however, find a significantly increased frequency of the broad specificity HLA- We have previously shown an increased frequency of HLA-DRw8 in American blacks with young-onset SLE (12). In the present study, we also found an increased frequency of HLA-DRw8 in younger black patients with SLE (Table 3); however, the difference did not reach statistical significance.…”

“…As has been shown previously (12,22), anti-U1 RNP and anti-Sm antibodies were found more frequently in American blacks than in Caucasians. A finding not reported previously, however, was that the significantly increased frequency of anti-RNP antibodies in the black patient population (when compared with age-matched Caucasian patients) was due to the marked increase among those <I8 years of age.…”

“…Reveille et al showed that early onset SLE patients ((20 years old) have a high frequency of HLA-DRw8, 21 and Manger et al reported that SLE patients with Fcc IIA-R/R131 and IIA/IIIA-R/R131, F/F158 polymorphisms have a younger onset of disease. 22 However, the number of early onset SLE subjects was very small (18 patients in the former study), or not mentioned.…”

“…HLA-DR and -DQ specificities were assigned to specific RFLP bands at the 10th International Histocompatibility Testing Workshop (24), as well as in our own (25,26) and others' studies (27-32). Specific DQA1 and DQB1 chain alleles in each individual were assigned by comparison with reported DQA1 and DQB1 RFLP's for specific HLA-D haplotypes as determined by the World Health Organization Nomenclature Committee for factors of the HLA system which met during the 7th International Congress of Immunology in 1989 (33).…”
