DLG-1 Is a MAGUK Similar to SAP97 and Is Required for Adherens Junction Formation

Abstract: Cellular junctions are critical for intercellular communication and for the assembly of cells into tissues. Cell junctions often consist of tight junctions, which form a permeability barrier and prevent the diffusion of lipids and proteins between cell compartments, and adherens junctions, which control the adhesion of cells and link cortical actin filaments to attachment sites on the plasma membrane. Proper tight junction formation and cell polarity require the function of membrane-associated guanylate kinase… Show more

“…Importantly, SAP97, cadherins, and integrins are all associated with the actin/spectrin cortical cytoskeleton. In fact, in non-neuronal cell systems, SAP97 is recruited to sites of cell-cell adhesion in a cadherin dependent manner and promotes cell-cell adhesion (Thomas et al, 1997;Firestein and Rongo, 2001).…”

“…First, BLJ mutant follicle cells decrease adhesion to each other, because as we have shown, they fail to form adherens junctions. Likewise in mammals, Dlg-1 has been shown to be required for adherens junction formation (Firestein and Rongo, 2001). Second, BLJ transformed cells develop novel adhesion interactions with germ cells, because they develop basolateral membrane proteins around their circumference, and because not only proteins of the adherens junction, but also those of BLJ (with the exception of Fas2) are present on germ cell membranes (data not shown).…”

“…Several of these targets are proteins found at regions of cell-cell contact, including the MAGI-1, -2 and -3 proteins Thomas et al, 2002) and MUPP1 , which are found at subapical tight junctions (Ide et al, 1999;Hamazaki et al, 2001), and the Dlg1 and Scribble proteins (Gardiol et al, 1999;Nakagawa and Huibregtse, 2000;Thomas et al, 2005), found at lateral adherens junctions (Bilder and Perrimon, 2000;Firestein and Rongo, 2001). This apparent level of redundancy is certainly a concern, but some of this is most likely a reflection of differences between different E6 proteins: targeting the same pathway but through different substrates.…”
