DJ-1 and androgen receptor immunohistochemical expression in prostatic carcinoma: A possible role in carcinogenesis

Abstract: AR nuclear expression was consistently present in benign and adenocarcinoma epithelium. But, there may be limited clinical use for AR expression in localized carcinoma due to its constant heterogeneity. DJ-1 with its oncogenic properties, specificity for prostatic carcinoma and homogenous expression gives an ideal complementary role to AR in the detection and treatment of prostatic carcinomas.

“…19) Increasing evidences pointed that AR was a critical mediator for anti-prostate cancer. [20][21][22] In view of this, researchers tended to explore agents to antagonize AR as improved therapeutic methods for prostatic cancer. A natural prenylflavonoid, also known as icaritin (ICT), inhibited AR-regulated genes in AR-positive prostate cancer cells via AR-depended pathways.…”

“…It is well accepted that DJ‐1 plays a critical role in the development of PCa mediated by AR signaling (Tillman et al, ; Osman et al, ), yet the detailed mechanism remains vague, which leaves a very interesting scientific question for the investigation. The effect of DJ‐1 on autophagic activity has been verified in other types of cell (Ren et al, ) but never in PCa.…”

“…Moreover, DJ‐1 binds AR to stimulate its transcriptional activity in androgen‐treated PCa cells, which contributes to the function of androgen and progression of PCa (Tillman et al, ). More important, the value of DJ‐1 immunohistochemical expression in the diagnosis of PCa has been proved (Osman et al, ). Not only is there a significant correlation of immunohistochemical scores between DJ‐1 and AR, but DJ‐1 is more specific than AR (Osman et al, ).…”

“…63) DJ-1 is translocated from the cytoplasm to nucleus upon addition of a mitogen to the culture medium 26) and is translocated to mitochondria after oxidative stress. 64) Increased levels of DJ-1 expression have been observed in various cancer cells and tissues, including breast, 51,52) prostate, 65,66) lung, 67,68) and endometrial, 69,70) thyroid, 71,72) pancreas, [73][74][75][76] esophageal, 77) ovary, 78,79) cervical, 80) liver, 81) gastric, 82 57,90) and leukemia. 91,92) Secretion of DJ-1 into the serum or nipple fluid has been found in patients with breast cancer.…”
