Diversity of Mycobacteriaceae from aquatic environment at the São Paulo Zoological Park Foundation in Brazil

Abstract: We investigated the species diversity of Mycobacteriaceae in surface water samples from six environments at the zoological park in São Paulo, Brazil. Three hundred and eighty isolates were cultivated and identified by phenotypic characteristics (growth rate and pigmentation) and sequencing of hsp65, rpoB and 16S rRNA genes. The results revealed that almost 48% of the isolates could be identified at the species level; about 50% were classified at the genus level, and only less than 2% of the isolates showed an … Show more

“…None of these organisms were found in water samples during our study so we were unable to evaluate their growth from environmental sources. However, at least some of these species including M. haemophilum [1], M. neoaurum [31], M. phlei [32] and M. xenopi [33] have been isolated from water samples by others. Only M. haemophilum and M. xenopi were noted by the CDC as environmental organisms of concern for HAIs.…”

“…In this sense, studies are essential for advancing our knowledge of microbial diversity, which has implications for fields as diverse as medicine, ecology, industry and environmental sciences. In our previous study, a large sampling of mycobacteria from water samples from the São Paulo Zoological Park was evaluated and the results suggested the presence of a putative new species ( Romagnoli et al, 2020 ). From these, five isolates were selected for further characterization regarding their taxonomic position.…”

“…Strains MYC017, MYC098, MYC101, MYC123 and MYC340 were isolated from sewage at the São Paulo Zoological Park in Brazil ( Romagnoli et al, 2020 ). Bacteria were cultivated in Middlebrook 7H10 medium supplemented with 0.5% glycerol and 10% OADC (oleic acid, albumin, dextrose, and catalase - Becton Dickinson, Franklin Lakes, USA) at 30°C for up to 10 days.…”

“…Strains MYC038, MYC040, MYC211, MYC221, and MYC223, identified as M. austroafricanum , were isolated from sewage and a lake at São Paulo Zoological Park Foundation in Brazil [ 33 ]. Bacteria were grown on solid Middlebrook 7H10 media supplemented with 10% OADC (oleic acid, albumin, dextrose, and catalase) (Becton Dickinson, Franklin Lakes, NJ, USA) at 30 °C for 5 days.…”
