Diversity of Mating-Type Chromosome Structures in the Yeast Zygosaccharomyces rouxii Caused by Ectopic Exchanges between MAT-Like Loci

Abstract: We investigated sex chromosome diversity in Zygosaccharomyces rouxii (Z. rouxii). In the current study, we show that the organization of the mating-type (MAT) locus is highly variable in the Z. rouxii population, indicating the MAT, HML, and HMR loci are translocation hotspots. Although NBRC1130 and CBS732 were originally two stocks of the type strain of the species, only NBRC1130 retains the original karyotype. A reciprocal translocation between the MAT and HMR loci appears to have occurred during the early p… Show more

“…S2) and placed the peak signal from chromosome 3 at position 897 kb, which is 3 kb from the leftmost IR and 5 kb from the MATalpha genes. The centromeric region is transcriptionally silent (SI Appendix, (3,4,(12)(13)(14)(15)(16)(17)(18). Saccharomycotina, Pezizomycotina, and Taphrinomycotina are subphyla within phylum Ascomycota (19).…”

“…Such events of unusual recombination can cause changes in breeding systems from heterothallism to homothallism as observed in Aspergillus nidulans (Paoletti et al , 2007), Neurospora spp. (Ellison et al , 2011; Gioti et al , 2012), S. sclerotiorum (Chitrampalam et al , 2013), Grosmannia clavigera (Tsui et al , 2013), and Zygosaccharomyces rouxii (Watanabe et al , 2013). Meiotic crossing-over events that reorganize the mating-type locus as a step toward homothallism likely occurred between the homologous chromosomes bearing the mating-type regions.…”

“…To summarize, the first round of long-range PCR was done with high-fidelity DNA polymerase (Phusion, Thermofisher) and the external primers 1, 2, 3, A, B, B′, C (Watanabe et al 2013), and DownMATa1R1 (this study) in 20 µL of reaction volume, following the manufacturer’s instructions. Subsequently, a seminested PCR amplification was done using a 1:20 dilution of the previous PCR and internal MTL locus-specific primers.…”
