Funds collection zebranie funduszy Summary Lyme disease, as one of tick-borne diseases, has been a current epidemiological problem in Poland and in the world for several years. The proportion of borreliosis infections caused by B. burgdorferi spirochaetes is increasing. Difficulties diagnosing this disease with conventional methods have led to growing molecular biology research aimed at developing improved diagnostic tools. Molecular biology methods include all techniques that allow the detection and analysis of nucleic acids. Among them there are methods based on PCR reaction, molecular typing methods (MLST), new generation sequencing (NGS). The current development of this field gives great research opportunities. With regard to B. burgdorferi spirochaete, molecular biology is used to: confirm Lyme borreliosis, identify and type Borreliella spirochaetes, detect them in tick vectors or intermediate hosts, as well as to identify co-infections between different Borreliella species and other tick-borne pathogens. They are meant to significantly improve diagnostic analyzes. This paper reviews the current methods used for the detection and identification of B. burgdorferi. Molecular mechanisms for the survival of spirochaetes in the host, infection vectors and clinical picture of Lyme disease were also discussed.

Barbour-Stoenner-Kelly II (BSK-II) and BSK-H media were used for cultivation and isolation of fastidious Borrelia species-the causative agents of Lyme borreliosis. Culture media have a limited shelf life and require adequate storage. Our goal was to assess how the growth of Borrelia would be affected by prolonged storage of media and inadequate storage conditions (BSK-H stored at +4°C for 2.5 years and BSK-II stored at À20°C for 11 years). Growth of different Borrelia afzelii, Borrelia garinii, Borrelia lusitaniae and Borrelia valaisiana strains was assessed during 2 weeks of incubation at 33°C. Monitored parameters included cell count per mL, morphology and motility. The results of this study have shown weaker growth of borrelia strains in BSK-H at +4°C (median final cell number of 1.5 9 10 6 /mL) than in BSK-II at À20°C (median final cell number of 7.75 9 10 6 /mL) and in fresh BSK-H media (median final cell number of 8.95 9 10 6 /mL). Duration of storage of media had no impact on Borrelia morphology and motility. Our results indicate that temperature of À20°C is optimal for long-term storage of medium, BSK-II stored for 11 years provided effective support to growth of Borrelia and may be employed for cultivation.

Increased prevalence of tick-borne disease in humans and animals is an important public health issue. Dogs can be clinically ill, reservoirs of tick-borne pathogens, or sentinel populations for research. Certainly, small-scale research has been conducted in dogs and this review will focus on the recent studies of emergent and re-emergent tick-borne pathogens in dogs in Serbia. Babesiosis stands out as the most common and important tick-borne disease in dogs. In addition to this pathogen, the presence of Hepatozoon canis and ?Candidatus Mycoplasma haematoparvum? in the blood of dogs was proved using molecular methods. Seroreactivity in dogs has been proven for Borrelia burgdorferi sensu lato (s.l.) complex, Ehrlichia canis, Ehrlichia ewingii, Anaplasma phagocytophilum, Anaplasma platys, Rickettsia conorii, and tick-borne encephalitis virus (TBEV). The presence of B. burgdorferi s.l. complex, A. phagocytophilum, Rickettsia spp. from the group of spotted fevers, and TBEV in ticks indicates the geographical distribution and potential for infecting humans, dogs, and other animals in Serbia. Further systematic, comprehensive, well designed, bacteriological, virological, parasitological, epidemiological, clinical (infectological), and acarological research is needed in Serbia.
