Previous difficulties in arthropod taxonomy (such as limitations in conventional morphological approaches, the possibility of cryptic species and a shortage of knowledgeable taxonomists) has been overcome by the powerful tool of DNA barcoding. This study presents a thorough analysis of DNA barcoding in regards to Pakistani arthropods, which were collected from Lahore’s Jinnah Garden. The 88 % (9,451) of the 10,792 specimens that were examined were able to generate DNA barcodes and 83% (8,974) of specimens were assigned 1,361 barcode index numbers (BINs). However, the success rate differed significantly between the orders of arthropods, from 77% for Thysanoptera to an astounding 93% for Diptera. Through morphological exams, DNA barcoding, and cross-referencing with the Barcode of Life Data system (BOLD), the Barcode Index Numbers (BINs) were assigned with a high degree of accuracy, both at the order (100%) and family (98%) levels. Though, identifications at the genus (37%) and species (15%) levels showed room for improvement. This underscores the ongoing need for enhancing and expanding the DNA barcode reference library. This study identified 324 genera and 191 species, underscoring the advantages of DNA barcoding over traditional morphological identification methods. Among the 17 arthropod orders identified, Coleoptera, Diptera, Hemiptera, Hymenoptera, and Lepidoptera from the class Insecta dominated, collectively constituting 94% of BINs. Expected malaise trap Arthropod fauna in Jinnah Garden could contain approximately 2,785 BINs according to Preston log-normal species distribution, yet the Chao-1 Index predicts 2,389.74 BINs. The Simpson Index of Diversity (1-D) is 0.989, signaling high species diversity, while the Shannon Index is 5.77, indicating significant species richness and evenness. These results demonstrated that in Pakistani arthropods, DNA barcoding and BOLD are an invaluable tool for improving taxonomic understanding and biodiversity assessment, opening the door for further eDNA and metabarcoding research.

Insect biodiversity drives ecological functioning and provides ecosystem services in different landscapes. Protected areas with the least anthropogenic disturbances provide resources with the least disturbance in habitat favouring insect species belonging to different guilds to coexist and contribute to the ecosystem processes. Studies have highlighted the significance of anthropogenic landscapes including agricultural ecosystems for harboring rich native biodiversity. To understand the species composition and abundance agricultural landscape, we explored insect fauna in Deva Vatala National Park (DVNP), Bhimber (Azad Kashmir), and associated croplands of Gujrat, Punjab, Pakistan. We aimed to assess the abundance, richness and diversity of the insect orders, families and species found in the two habitats i.e. national park and croplands. Fortnightly surveys were conducted during 2017-2019. Sampling methodology consisted of handpicking, using sweep nets, light traps, pitfall traps and insect collection was identified by using taxonomic keys. A total of 239 insect species representing 10 orders and 69 families were collected and identified from both habitat types. For both diversity and abundance, we detected a significant difference in community composition at species, family and order levels. Several insect orders and families demonstrated significant differences (p< 0.05) in either abundance or richness or both between the two types of habitats. Several families of Coleoptera, which contains dung beetles, ground beetles, ladybird beetles showed greater richness in DVNP. However, we also detected that several insect orders showed greater richness in the croplands of Gujrat. But we observed the pattern of the greater abundance of insect orders in DVNP. Both types of habitat can have important consequences on insect fauna having great importance in the ecosystem services and functions. Protected areas nearby associated croplands have the potential to contribute immensely to biodiversity conservation and ecosystem service provision both at the local scale and the landscape scale.

Arboreal Tiger Beetles Recorded from Lowland Crop Cultivations in Sri Lanka

Purpose : Thirty-one species of arboreal tiger beetles are known from Sri Lanka of which 25 species are endemic. However, their habitat types are poorly documented and the available records are far outdated. Therefore, a survey of tiger beetles was carried out to determine their present occurrence with emphasis on agricultural habitat types. Research Method : Forty-six locations of the country, covering eighteen districts, all provinces, representing a majority of bioclimatic zones except those in Montane Sri Lanka were surveyed for arboreal tiger beetles. Sampling was conducted using the visual encounter method. Collected beetles were identified using taxonomic keys and descriptions. Findings : Eight species of arboreal tiger beetles were collected from the survey. Majority of the species (06) were collected from crop cultivations of coconut and also from tea, fruit farms, betel leaf, cinnamon and pepper. Four species of Derocrania and two species of Tricondyla were recorded from the cultivations and all had fused elytra and hence unable to fly. Derocrania scitiscabra was the dominant arboreal tiger beetle species in the crop cultivations. Originality/ Value : The study documents hitherto unrecorded habitat types for a poorly documented important beetle group of Sri Lanka. It further provides information for future research on the possibility of using arboreal tiger beetles as bio-control agents of insect pests of agricultural crops.
