Objective: The objective of this study was to investigate the group/subgroup of phytoplasma agent in peppers showing phytoplasma symptoms. Material and Methods: In this study, plants collected from Iğdır province in 2020 were analyzed using direct and nested PCR tests, and BLASTn, iphyClassifier, Mega 7, and pDRAW32 programs were used. Results: In the tests performed, approximately 1.2 kb of DNA fragments specific to phytoplasma were obtained. The 16S rRNA nucleotide sequence (1254 bp in length) (OM663745) revealed that it was showed more than 99.44% nucleotide similarity to other ‘Ca. P. trifolii’ members. The tentative RFLP and phylogenetic analyzes performed proved the ‘Ca. P. trifolii’ the infection from the Clover proliferation group (16SrVI) group and subgroup A in symptomatic pepper plants. Conclusion: The presence of ‘Ca. P. trifolii’ in naturally infected peppers in Iğdır province of Turkey was detected using PCR-RFLP and cladistic analysis.

Following a request from the European Commission, the EFSA Panel on Plant Health performed a pest categorisation of four phytoplasmas of tuber‐forming Solanum spp. known to occur only outside the EU or having a limited presence in the EU . The only tuber‐forming species of Solanum reported to be phytoplasma infected is S. tuberosum . This opinion covers ‘ Candidatus Phytoplasma americanum’, ‘ Ca . P. aurantifolia’‐related strains ( GD 32; St_ JO _10, 14, 17; PPT ‐ SA ; Rus‐343F; PPT ‐ GTO 29, ‐ GTO 30, ‐ SINTV ; Potato Huayao Survey 2; Potato hair sprouts), ‘ Ca . P. fragariae’‐related strains ( YN ‐169, YN ‐10G) and ‘ Ca . P. pruni’‐related strains (Clover yellow edge; Potato purple top AK pot7, MT 117, AK pot6; PPT ‐ COAHP , ‐ GTOP ). Phytoplasmas can be detected by molecular methods and are efficiently transmitted by vegetative propagation. Phytoplasmas are also transmitted in a persistent and propagative manner by some insects belonging to families within Cicadomorpha, Fulgoromorpha and Sternorrhyncha (order Hemiptera). No transovarial, pollen or seed transmission has been reported. The reported natural host range of the phytoplasmas categorised here varies from restricted (‘ Ca . P. americanum’, and ‘ Ca . P. fragariae’‐related strains) to wide (‘ Ca . P. aurantifolia’‐related strains and ‘ Ca . P. pruni’‐related strains), thus increasing the possible entry pathways in the latter case. S. tuberosum is widely cultivated in the EU . All the categorised phytoplasmas can enter and spread through the trade of host plants for planting, and by vectors. Establishment of these phytoplasmas is not expected to be limited by EU environmental conditions. The introduction of these phytoplasmas in the EU would have an economic impact. There are measures to reduce the risk of entry, establishment, spread and impact. Uncertainties result from limited information on distribution, biology and epidemiology. All the phytoplasmas categorised here meet the criteria evaluated by EFSA to qualify as potential Union quarantine pests, and they do not meet all the criteria to qualify as potential regulated non‐quarantine pests, because they do not occur or are not known to be widespread in the EU . ...

Фитоплазменные болезни были известны задолго до открытия их возбудителя. С начала 1930-х годов на постсоветском пространстве была признана инфекционная природа заболевания, известного под названием «столбур томатов». Предпринимались попытки найти переносчика этого заболевания (И.К. Корачевский, 1934; В.Л. Рыжков с соавт., 1934). В 1945 году был обнаружен вектор болезни-циксиида Hyalesthes obsoletus Sign. (К. Сухов с соавт., 1946). В течение нескольких десятилетий в нашей стране и за рубежом безуспешно велись поиски возбудителя инфекционных болезней растений с симптомами карликовости, пожелтения, изменений в листьях и генеративных органах, не способного расти на искусственных питательных средах. Открытие было сделано только в 1967 году японскими исследователями (Y. Doi с соавт., 1967). Возбудителем заболевания оказался ранее неизвестный фитопатоген из класса Molliсutes, сходный с патогенами микоплазмы животных. Позже было установлено, что жизненный цикл фитоплазм связан как с клетками флоэмы растений, в которых они размножаются, так и с насекомыми-переносчиками из отряда Hemiptera, которые питаются соком растений, а также способны поддерживать размножение фитоплазм в своем организме и даже передавать их потомству. Фитоплазмы, как и другие Molliсutes, не имеют клеточной стенки. Они обладают минимальным геномом, известным среди клеточных организмов, что обусловливает их обязательное паразитическое существование. В бывшем СССР фитоплазменные болезни широко изучались во Всесоюзном научноисследовательском институте защиты растений и в других учреждениях. Для этого использовали методы электронной микроскопии, иммунологические исследования, осуществляли успешное культивирование патогена на искусственных питательных средах. Спустя три десятилетия итальянские ученые смогли добиться культивирования инфекционного агента на искусственных средах и подтвердили его принадлежность к фитоплазме, секвенировав ДНК (А. Bertaccini с соавт., 2010; N. Contaldo с соавт., 2012, 2013). В 1990-е годы с внедрением молекулярно-генетических методов был сделан шаг вперед в области диагностики и изучения свойств фитоплазм. Их таксономия была разработана на основе консервативного гена 16S рибосомальной РНК и получила дальнейшее развитие с привлечением других консервативных и менее консервативных генов (I.-M. Lee с соавт.
