Diurnal Fluctuations of Nitrate Uptake and In Vivo Nitrate Reductase Activity in Pima and Acala Cotton

Abstract: This study was conducted to determine whether diurnal fluctuations of nitrate reductase activity in two cotton species, Pima (Gossypium barbadense L.) and Acala (Gossypium hirsutum L.), are regulated by NO−3 uptake or leaf NO−3 concentration. The two species differ greatly in their N use efficiency. The seedlings were grown for 25 d in nutrient solutions containing 0.05, 0.10 or 1.0 mM NO−3 under a 14‐h light/10‐h dark cycle at 30°C/20°C. Uptake rates were measured by following NO−3 depletion from the uptake s… Show more

“…Therefore, we conclude that photosynthetic products play an important role in NO 3 -N absorption over a longer period. Previous studies have assumed that photosynthetic products are involved in the NO 3 -N absorption via nitrate assimilation process (Aslam et al, 1979(Aslam et al, , 2001Cárdenas-Navarro et al, 1998;Dechorgnat et al, 2011;Delhon et al, 1996;Li et al, 2013;Rideout and Raper Jnr, 1994). Hence, the factors affecting NO 3 -N uptake may include transpiration and plant photosynthesis.…”

“…NR, the first key enzyme of NO 3 ( assimilation, can regulate plant N nutrition and affects plant physiology including photosynthesis and carbon assimilation. NR activity may vary in different plant organs and developmental stages (Sun and Yan 2004) and show diurnal variation rhythm with the maximum activity occurring around mid day (Aslam et al 2001;Sun and Yan 2004). Leaf NRA did not show any obvious correlation with leaf area and/or lamina mass per unit area in Camptotheca accuminata seedlings (Sun and Yan 2004).…”

“…After 6 d, the seedlings were transferred to a stainless steel screen (mesh size 5 mm × 5 mm) suspended ≈5 cm above the surface of 5 L aerated N‐free, one‐quarter‐strength Hoagland solution (Hoagland and Arnon, 1950) contained in a plastic beaker. To avoid diurnal fluctuations in NO − 3 uptake due to light/ dark transitions and temperature (Aslam et al, 2001), the seedlings were transferred into a growth chamber (Western Environmental, Napa, CA) set for continuous light and 28°C with 60 to 65% relative humidity. Light was supplied by metal halide and incandescent lamps.…”
