Diterpenoid Alkaloids from the Aerial Parts of Aconitum flavum Hand.-Mazz

Abstract: Sixteen diterpenoid alkaloids (DAs), including six aconitine-type alkaloids (5 and 9 − 13), seven 7,17-seco-aconitine-type alkaloids (1 − 4, 6 − 8), two napelline-type alkaloids (14 and 15) as well as one veatchine-type alkaloid (16), were isolated from the aerial parts of Aconitum flavum Hand.-Mazz. In which, flavumolines A − D (1 − 4) were four new ones, and flavumoline E (5) was reported as natural compound for the first time. Their chemical structures were elucidated by the analysis of extensive spectrosco… Show more

“…The AFHM total alkaloids were detected by HPLC/QTOF-MS ( Figure 1 ), and the mass/charge ratio and molecular weight of the detected components were compared with the database of traditional Chinese medicine. There were 11 alkaloids ( Table 1 ), namely 6-O-demethylneoline, songorine, 12-epi-napelline, neoline, 10-hydroxyneoline, jesaconine ( 25 ), benzoylaconitin, 3-deoxyaconitine ( 26 ), mesaconitine, acetylaconitine, and aconitine ( 27 ), which were consistent with existing research reports. The contents of aconitine and 3-deoxyaconitine in total alkaloids were determined by HPLC.…”

“…In our laboratory, genomic DNA was extracted using the modified CTAB method [ 55 , 56 ] in 2021 and 2022. The concentration and purity of the extracted DNA were determined by spectrophotometer using NanoDrop 2000c (Thermo Scientific, Waltham, MA, USA) equipment and 0.8% agarose gel.…”

“…Franchetine-type C19-DAs are distinguished from aconitines by an additional ether between C-6 and C-17. Ten novel alkaloids (159-168) are classified as franchetine-type C19-DAs [43,56,76]. The plant sources are shown in Table 7.…”

“…Detailed chemical structures are shown in Figure 7. Compounds 159, 161-166, and 168 have a double bond between C-7 and C-8, whereas compounds 160 and 167 have a 7,8-epoxy unit [43,56,76]. In addition, Compounds 161 and 163 have a characteristic hydroxyl group at C-16 instead of the methoxy group joint in DAs.…”
