Distribution of Radiolabelled Lymph Cells in Lymph Nodes and the Migratory Properties of Blood Lymphocytes in Sheep

Abstract: The results found in this study supported the concept of lymphocyte populations with preferential migratory pathways. Preferential localization of lymphocytes in lymph nodes could not explain the non-random lymphocyte migration patterns observed in sheep. The migration of lymphocytes isolated from the blood was similar to that of lymphocytes in the efferent lymph of a subcutaneous lymph node, but was different from the migration of lymphocytes isolated from efferent intestinal lymph. The subcutaneous lymph nod… Show more

“…Early studies of lymphocyte recirculation in rats were performed using cells collected by thoracic duct cannulation (Gowans and Knight, 1984). Subsequent studies in sheep used cells collected from thoracic duct or efferent lymph from individually cannulated lymph nodes (Hall et al, 1977;Chin and Hay, 1984). A number of studies of lymphocyte recirculation in rodents used cells obtained by excision of lymph nodes and in vitro preparation of cell suspensions that are then labeled by various methods and injected into syngeneic recipients (Griscelli et al, 1969;Freitas and De Sousa, 1975;Roux et al, 1977;Rose et al, 1978).…”

“…Also, little is known about the effect of blood lymphocyte purification and labeling procedures on the ability of the cells to recirculate (10). Second, the blood contains a composite of lymphocytes from many sources.…”

“…Historically, ex vivo radionucleotide labelling of lymphocytes was utilized to track their anatomical distribution upon adoptive transfer in vivo by autoradiography of histological sections (Chin and Hay, 1984;Gowans and Knight, 1964;Hall et al, 1976;Rannie et al, 1977). This approach was crucial in providing a general overview of the localization of the cells of interest, but the biohazardous nature of radioisotopes, the transfer of radioisotope from labelled cells to bystanders and loss of signal due to radioactive decay hampered its utility for long term monitoring host immune responses in vivo (Parish, 1999).…”
