Plum pox virus (PPV) is the causal agent of Sharka disease. Since its discovery, Sharka has been considered as a calamity in plum orchards. PPV is present worldwide in many Prunus species, causing great economic losses. In highly susceptible plum varieties, such as Pozegaca, PPV causes a premature fruit drop and reduces fruit quality, which leads to total yield loss. Eight PPV strains (PPV-M, PPV-D, PPV-EA, PPV-C, PPV-Rec, PPV-W, PPV-T and PPVCR) have been recognized so far. Three major strains (PPV-M, PPV-D and PPV-Rec) are the most widely dispersed and occur frequently in many European countries. Other strains are of minor importance due to their limited host preferences or geographic distribution. So far, all three major strains have been identified in Serbia. Evaluation of the presence and concentration of PPV (Plum pox virus) in selected rootstocks was carried out in 2016 - 2017. For the purpose of the experiment we used rootstocks derived from crossbreeding of Prunus davidiana (Carr.) Franch, such as Cadaman and Barrier, and also a P. davidiana seedling. Peach seedling rootstock GF-305 was used as a control. The rootstocks were inoculated artificially with PPV strain M (Marcus). Both the rootstock and the inoculum were tested for presence of the virus by a serological method -semiquantitative DAS-ELISA test and molecular methods -RT-PCR, real-time RT-PCR and RT-LAMP. During the growing season the plants were evaluated for symptom intensity by using a scoring scale. The results show interdependency between symptom intensity and the amount of PPV in leaves, with DAS-ELISA test giving less positive samples than RT-PCR. The RT-LAMP and real-time RT-PCR methods were capable of revealing low concentrations of the virus even in symptom-free plants. The lowest PPV concentrations of all the four rootstocks were detected by real-time RT-PCR in P. davidiana. The highest PPV concentrations were detected in Barrier rootstock. In inocula, the lowest concentration was found in the inocula on Cadaman rootstock, whereas the highest PPV concentration was detected in the inocula inoculated on Barrier rootstock.

“…Currently, the offi cial sources have reported the PPV spread in peach orchards through the Czech Republic (Polák, 2004) as well as in other EU countries. Moreover, PPV virus is widely spread in natural environment of the Czech Republic (Polák, 2002;Polák and Komínek, 2009) so it is unlikely to guarantee a PPV-free area (Hnízdil, 2010). Finding a source of immunity or resistance within genus Prunus persica L., or in the wide gene pool in various geographical groups seems impossibly, the strategy to breed PPV resistant peaches gets even more complicated.…”

The degree of PPV occurrence was monitored in a young peach orchard during 2010–2011. This study evaluates the percentage of infected trees in 2010 as well as the intensity of PPV symptoms in 2011 in two peach varieties „Royal Glory“ and „Symphony“ grafted on seven different rootstocks of Prunus species after the natural infection. The different intensity of PPV symptoms was proved on infected parts such as flowers, leaves, fruits and from rootstocks’ point of view.In 2010 were PPV symptoms most often detected in variety ’Royal Glory’ grafted on rootstock Julior where 100% of trees were infected. No visible symptoms were observed in trees of variety ’Symphony’ on Pumiselect rootstock. As for the rootstocks evaluation, PPV symptoms were most numerous in trees grafted on MRS 2/5 rootstock (94.78%), at least were monitored on Lesiberian rootstock (28.47%).The intensity of PPV symptoms on flowers, leaves and fruits in each combination was evaluated in 2011. Generally, the PPV symptoms were present mainly on flowers and less on fruits.
