Distribution of cytomegalovirus genotypes among ulcerative colitis patients in Okinawa, Japan

Abstract: Background/AimsTo determine the prevalence of glycoprotein B (gB), glycoprotein N (gN), and glycoprotein H (gH) genotypes of human cytomegalovirus (HCMV) superimposed on ulcerative colitis (UC) patients in Japan.MethodsFour archived stool samples and 7-archived extracted DNA from stool samples of 11 UC patients with positive multiplex polymerase chain reaction (PCR) results for HCMV were used UL55 gene encoding gB, UL73 gene encoding gN, and UL75 gene encoding gH were identified by PCR. Genotypes of gB and gly… Show more

“…As the patient can be infected by several genotypes, some of them could be associated to the severity of the disease. Only a few studies have depicted such relation [ 93 , 94 ]; more data are needed, especially around the world as some genotypes are found mainly in Asian countries.…”

“…Detection of CMV DNA in stool specimens could also be an alternative to biopsy examination [ 94 , 100 , 101 , 102 , 103 ]; but, to date, the sensitivity of the assays is too low, leading to false-negative results, even in allograft patients with high viral load in the tissue [ 104 ].…”

“…Genotyping of CMV has mainly focused on gB, gN, and gH, which play a role in virus entry and may influence the infectivity or pathogenicity of CMV. 1 , 2 It has been hypothesized that genetic variation among CMV strains may underlie strain-specific clinical manifestations. Our previous research revealed that there might be a potential association between the genotypes of CMV and neonatal thrombocytopenia, and the detection of some specific genotypes might be indicative of severe manifestations in infants with CMV infection.…”

“…1 Several envelope glycoproteins, encoded by UL55, UL73, and UL75, including glycoprotein B (gB), glycoprotein N (gN), and glycoprotein H (gH), respectively, have been evaluated in clinical CMV iso-lates because of their influence on tissue tropism and virulence. [2][3][4] The UL144 polymorphisms are concentrated in the 5′ end of the gene, especially in cysteine-rich domain 1 (CRD1), that may bind the B-and T-lymphocyte attenuator proteins. This domain's activity may vary among polymorphisms, leading to a range of clinical symptoms and prognoses.…”
