Despite the availability of anticancer drugs, breast cancer remains the most death-causing tumor-related disease in women. Hence, there is a need for discovery and development of efficient alternative drugs, and sources such as plants need to be explored. In this study, antioxidant capacities and inhibitory effects against MCF7 cells of the extracts of stem bark of three Nigerian medicinal plants (Detarium microcarpum, Guiera senegalensis, and Cassia siamea) were investigated. The D. microcarpum extracts had the highest antioxidant and antiproliferative effects, followed by that of G. senegalensis, and the C. siamea extracts had minimal effects. The IC50 values of the methanol and aqueous extracts from the three plants that inhibited the proliferation of MCF7 cells ranged from 78–> 500 μg/ml. Moreover, all the plant extracts but the aqueous extract of Cassia siamea exhibited antimetastatic action and induced apoptosis and cell cycle arrest in MCF7 cells. Liquid chromatography/time-of-flight/mass spectrometry profiling revealed that the five potent extracts contain many phenols and omega-6 fatty acids, and some of the identified compounds (isorhamnetin, eupatorin, alpinumisoflavone, procyanidin B3, syringin, and gallic acid) have been reported to have antiproliferative effects on cancer cells. Hence, the stem bark of these plants could be potential sources of antibreast cancer agents.

De novo Assembly of Leaf Transcriptome in the Medicinal Plant Andrographis paniculata

Andrographis paniculata is an important medicinal plant containing various bioactive terpenoids and flavonoids. Despite its importance in herbal medicine, no ready-to-use transcript sequence information of this plant is made available in the public data base, this study mainly deals with the sequencing of RNA from A. paniculata leaf using Illumina HiSeq™ 2000 platform followed by the de novo transcriptome assembly. A total of 189.22 million high quality paired reads were generated and 1,70,724 transcripts were predicted in the primary assembly. Secondary assembly generated a transcriptome size of ~88 Mb with 83,800 clustered transcripts. Based on the similarity searches against plant non-redundant protein database, gene ontology, and eukaryotic orthologous groups, 49,363 transcripts were annotated constituting upto 58.91% of the identified unigenes. Annotation of transcripts—using kyoto encyclopedia of genes and genomes database—revealed 5606 transcripts plausibly involved in 140 pathways including biosynthesis of terpenoids and other secondary metabolites. Transcription factor analysis showed 6767 unique transcripts belonging to 97 different transcription factor families. A total number of 124 CYP450 transcripts belonging to seven divergent clans have been identified. Transcriptome revealed 146 different transcripts coding for enzymes involved in the biosynthesis of terpenoids of which 35 contained terpene synthase motifs. This study also revealed 32,341 simple sequence repeats (SSRs) in 23,168 transcripts. Assembled sequences of transcriptome of A. paniculata generated in this study are made available, for the first time, in the TSA database, which provides useful information for functional and comparative genomic analysis besides identification of key enzymes involved in the various pathways of secondary metabolism.

Andrographis paniculata (Burm. f.) Wa ll. ex Nees (Acanthaceae) with immense medicinal importance lacks information on its biosynthetic pathway genes and their regulatory role in the production of pharmaceutically important andrographolide. Copalyl diphosphate synthase (CPS) is involved in the production of copalyl diphosphate, a precursor for many bioactive compounds with particular reference to diterpene lactone. In this study, we elucidated the structural and functional aspects of A. paniculata CPS (ApCPS). Composition of amino acids and hydrophobic nature of ApCPS were analysed and identified as non trans-membrane protein. A chloroplast transit peptide and mitochondrial targeting peptide in ApCPS were identified. Protein secondary structure prediction has given insight on the distribution of helix (52.52%), loop (45.91%) and strands (1.56%) in ApCPS. The homology modelling of ApCPS was carried out with SWISS MODEL. The validation of 3D model using PROCHECK revealed that 91.74% of the residues have averaged 3D-1D score >= 0.2 which is structurally reliable. In Ramachandran plot, 90.9% amino acid residues were found in most favoured region. Phylogenetic tree was constructed using MEGA 7.0 by taking eudicots, monocots, gymnosperms and fungal species. Among them, ApCPS was clustered within eudicots and closely related to Sesumum indicum in Laminales. Protein-protein interaction study using STRING10 revealed that CPS interacts with gibberilic acid and terpene synthase related proteins. In Arabidopsis thaliana, CPS coexpression was seen with gibberelic acid related proteins. The present in silico analysis will be useful in understanding the structural, functional and evolutionary diversification of ApCPS.
