The Red-headed Wood Pigeon Columba janthina nitens is endemic to the Ogasawara Islands, an oceanic island chain located 1000 km south of the main islands of Japan. The subspecies is at high risk of extinction because of its small population size and restricted habitat range. We undertook genetic analyses of this pigeon using sequences of a portion of the mitochondrial control region and five microsatellite markers to estimate the genetic characteristics of two wild populations from the Bonin and Volcano Islands, as well as one captive breeding population. The genetic diversity of the wild individuals was exceptionally low in both the mitochondria (nucleotide diversity = 0.00105) and at the microsatellite (3.2 alleles per locus and H E = 0.12) loci. Higher numbers of microsatellite genotypes were observed in the Volcano Islands population than in the Bonin Islands population, which may be because of the relatively low impact of human disturbance. The most common mitochondrial haplotypes and microsatellite alleles observed in the two wild populations were completely fixed in the captive population. Our results suggest that the genetic diversity of the captive population needs to be increased. However, introduction of a wild individual into a captive population can lead to a decreased genetic diversity in the wild population and therefore should be done with caution. The genetic differentiation between the Bonin and the Volcano island groups was low, and the populations of the two island groups should be regarded as a single evolutionarily significant unit. However, special consideration is required for habitat conservation in the Volcano Islands, which may be functioning as a sanctuary for the Red-headed Wood Pigeon. For the long-term conservation of threatened bird species that live on remote oceanic islands, determination of management units considering gene flow caused by their flying capacity and maintenance of genetically suitable wild and captive populations are essential.

Summary:The Eurasian Collared Dove Streptopelia decaocto is one of the most successfuly expanding vertebrate species in the world; therefore, it has attracted special scientific focus. This review summarises the species' achievements related to its rapid expansion and the ecological consequences with regard to the population genetic assessments. It also highlights the expansion dynamics of the species and the ecosystems to which it is most adapted. Finally, the effects of this species on the newly colonised ecosystems are illustrated. The study of the genetic structure between populations to make inferences about expansion dynamics has been neglected so far. Thus, we survey the opportunities of using molecular markers for genetic analysis in populations of Eurasian Collared Dove as a possible means for future studies. This work is important given the lack of information on the mentioned fields, and it calls attention to the importance of further interdisciplinary studies from different scientific fields using this species.

As of 2014, 89 species have been legally designated as "nationally endangered species of wild fauna and flora," including 37 avian species in Japan. To facilitate the breeding of endangered species and the maintenance of protected habitats, a program for the rehabilitation of natural habitats and maintenance of viable populations is being established by the Ministry of the Environment and other related ministries for 49 of the species on the above list. The programs have already been established for 15 such avian species, and are currently managed by zoos or local authorities. Maintaining genetic diversity is necessary to ensure the stable procreation of endangered species. A decline in breeding potential due to inbreeding can be prevented by careful maintenance of genetic diversity. In addition to the prevention of inbreeding depression, the maintenance of genetic diversity safeguards species' resistance to pathogens, as well as their capacity to cope with environmental change. This manuscript reports on the status of currently available genetic information for nationally endangered avian species in Japan, with a particular focus on the 15 species included in the conservation and propagation program. In the NCBI "Nucleotide" database, 315 entries were related to sequence information from the 15 endangered species. Mitochondrial DNA-related sequences constituted 238 (75.6%) of these entries. In the NCBI "Genome" database, genome sequence entries for three species were found. As is the case with mitochondrial DNA, microsatellite loci are very useful for endangered species conservation to evaluate genetic diversity. However, information about the endangered avian species' microsatellite makers is limited. This situation could have a considerable negative impact on captive breeding projects in particular. Hence, the development of microsatellite markers for these species is a priority.
