Distinct roles for PP1 and PP2A in the Neurospora circadian clock

Abstract: Phosphorylation of the Neurospora circadian clock protein FREQUENCY by several kinases promotes its degradation and is important for the function of the circadian feedback loop. Here, we show that FRQ is less stable in a ppp-1 (catalytic subunit of PP1) mutant, resulting in its advanced phase and short period. In contrast, FRQ stability is not altered in a rgb-1 (a regulatory subunit of PP2A) mutant, but levels of frq protein and mRNA are low, resulting in a low-amplitude and longperiod oscillation of the cloc… Show more

“…A strain lacking functional RGB-1 (rgb-1 RIP ), a regulatory subunit of PP2A, is severely compromised in cell growth and displays irregular conidiation patterns with >24-h rhythms. Although FRQ stability is not altered, overall levels of frq RNA and FRQ protein are reduced in the rgb-1 RIP strain (Yang et al 2004). The low expression of frq RNA reflects a function of PP2A in activating WCC, the transcriptional activator of frq .…”

“…FRQ is dephosphorylated by PP1 and PP2A in vitro, and both phosphatases regulate FRQ and the circadian clock in vivo in a specific manner (Yang et al 2004). It is not known whether these phosphatases are expressed in rhythmic fashion.…”

“…We estimate that ∼40%-50% of WC-1 is degraded through this process. Phosphorylation-dependent protein degradation has been demonstrated for FRQ in Neurospora, a critical process regulating the circadian clock (Liu et al 2000;Gorl et al 2001;Yang et al 2002Yang et al , 2004He et al 2003).…”

“…Under normal physiological conditions, FRQ is phosphorylated by CK-1a, CKII, and PKA (21,22,(25)(26)(27)(28). On the other hand, FRQ is also dephosphorylated and stabilized by protein phosphatases PP1 and PP4 (20,29). In the ck-1a, cka, and ckb-1 mutants, FRQ is hypophosphorylated and more stable than in the wild-type strain, leading to arrhythmic or long period conidiation rhythms in constant darkness (22,26,27).…”
