Distinct cardioprotective effects of 17β-estradiol and dehydroepiandrosterone on pressure overload–induced hypertrophy in ovariectomized female rats

Abstract: Taken together, these results from an ovariectomized rat model of PO-induced cardiac dysfunction show that DHEA but not E2 elicits a cardioprotective action through σ1 receptor activation. DHEA-induced Akt-eNOS activation through σ1 receptors is probably associated with its cardioprotective activity.

“…After immunoprecipitation with an anti-IP 3 R3 antibody, immunoprecipitants were separated on SDS-PAGE and subjected to Western blot analysis using an anti-DsRed antibody. In immunoblotting analyses to determine protein levels, cultured Neuro2a cells were washed with PBS at 4°C and stored at −80°C until analyses were performed [23]. For assays, frozen Neuro2A cells were homogenized using methods previously described [18].…”

“…Immunoprecipitation and Western blotting were performed using previously described methods [18,23]. Immunoprecipitation analysis of σ 1 R and IP 3 R3 was undertaken using lysates of Neuro2A cells with or without transfection with σ 1 R-mCherry or σ 1 R E102Q -mCherry.…”

“…Although these mechanisms remain unclear, our work suggests that the cardiac sigma-1 receptor and its downstream signaling protect the heart against TAC (22) and PO-induced cardiomyopathy (21,25). Previously, we also reported in several studies that the neurosteroid dehydroepiandrosterone (DHEA) serves as an endogenous ligand for the sigma-1 receptor and that treatment with DHEA ameliorated PO-induced cardiac injury by up-regulating the cardiac sigma-1 receptor in heart and activating downstream signaling (21,26,27). In support of our hypothesis, a recent study demonstrated significant reduction in DHEA-sulfate concentrations in brain circumventricular and hypothalamic tissue after aortic banding (23).…”
