Dissolvable Immunomodulatory Microneedles for Treatment of Skin Wounds

Pejman Ghelich,
Mohamadmahdi Samandari,
Alireza Hassani Najafabadi
et al.

Abstract: Sustained inflammation can halt or delay wound healing, and macrophages play a central role in wound healing. Inflammatory macrophages are responsible for the removal of pathogens, debris, and neutrophils, while anti‐inflammatory macrophages stimulate various regenerative processes. Recombinant human Proteoglycan 4 (rhPRG4) has been shown to modulate macrophage polarization and to prevent fibrosis and scarring in ear wound healing. Here, we engineered dissolvable microneedle arrays (MNAs) carrying rhPRG4 for t… Show more

“…TNFα and IL1β, known as pro-inflammatory cytokines, demonstrated overexpression in the presence of DLP hydrogels and PRP at 24 h, indicating their role in initiating the macrophage early pro-inflammatory response. The immediate pro-inflammatory response by macrophages in the presence of foreign material is a highly expected and well-known fact . The subsequent return of these pro-inflammatory marker expressions in treated cells to normal levels in reference to untreated cells at 72 h suggests a controlled resolution of the pro-inflammatory phase (Figure B,C).…”
