Dissolution behavior of Olmesartan Medoxomil drug in Polymeric Micelles of Soluplus and Pluronic F127

Miconazole nitrate (MN) is a broad-spectrum antifungal agent which suffers poor solubility and impermeability to the ocular tissue which limits its use in the treatment of ocular infections especially fungal keratitis, which is considered one of the most prevailing ocular infections. The current study aims to utilize polymeric mixed nano-micelles for the ocular delivery of MN using 33 full factorial design by varying the ratios of Pluronic® P123, Pluronic® F127, and Tetronic® T701 while the monitored responses were particle size, cloud point (CP), encapsulation efficiency (%EE) and %released at 6 hours (%Q6) at pH 7.4. The optimized formula was incorporated with Soluplus® (SP) to further enhance the (%EE) and the resultant formula was assessed in vitro as well as in vivo against C. albicans in treatment of induced ocular candidiasis using rabbits as a model animal. Results revealed that the optimized formula which comprised F127 and P123 in a ratio of (2:1) when incorporated with SP (SP-MPM) resulted in an increase in %EE from 35.12±3.18 to 99.19±7.03 with a particle size of 44.39±2.68nm with a sustained release profile and stability for 3 months at 4±2°C. In vivo results demonstrated the enhanced ability of SP-MPM for treatment of ocular candidiasis with enhanced % inhibition and susceptibility to C. albicans compared to 0.2% MN suspension confirmed with histopathological examination of rabbit’s eyes after 7 days of treatment with the absence of any degenerative effect to the ocular tissue. Thus, it can be concluded that SP-pluronic mixed nano-micelles offer a successful and stable ocular delivery platform for antifungal drug MN ensuring both its safety and efficacy.

Dissolution of drugs is a prerequisite in the development of their oral dosage forms with satisfactory bioavailability. In this investigation, solid dispersion of the model drug indomethacin in gelucire (50/13) and inclusion complexation in sulfobutyl ether beta cyclodextrin are employed to enhance the dissolution. Fast dissolving products (F1-F8) of indomethacin are prepared by different methods such as solvent evaporation, freeze drying and kneading and comparatively evaluated. The ternary and binary systems of the products are prepared and evaluated for their efficiency in increasing the dissolution. The dissolution efficiency values (DE30) were found to be higher for solid dispersions (73.34%) and inclusion complexes (53.12%) than the pure drug indomethacin (3.32%). The x ray diffraction and differential scanning calorimetry studies revealed that the crystalline drug is converted into an amorphous form in the dispersions and inclusion complexes. Soluplus and gelucire are employed as ternary components in the solid dispersions and inclusion complexes respectively and are found to be resulting in ternary systems which were found to be superior to the binary systems. A 22 full factorial experimental design in order to investigate the combined influence of method of preparation and nature of the product. It was observed that the method of preparation and nature of the product (a ternary system or a binary system) will influence the extent of improvement of dissolution of the drug.

Background and objective: Glycyrrhiza glabra is one of the useful medicinal plants with increasing demand for (herbal medicines, health products, pharmaceuticals). Glycyrrhiza glabra used in traditional medicine across the world for its ethnopharmacological value. Materials and Method: this study was included the evaluation of the effects of green synthetic nanoparticles of Glycyrrhiza glabra aqueous extract loaded with silver nitrate on antibacterial activity and anti-inflammatory activity through burn healing effect. The antibacterial activity of green synthetic silver nanoparticles was studied against one type of Gram-negative (E.coli) and Gram-positive (staphylococcus aureus). Also, the effects of burns healing effect of green synthetic silver nanoparticle was evaluated using albino male mice. The activity of green synthetic nanoparticle at (1.5mM) was investigated in compared with sliver sulfadiazine as positive control and negative control mice (without any treatment) by determining days require for healing. Results: the result showed that different nanoparticles concentrations (1, 1.5, 1.75, 2 mM) can inhibit the bacterial isolate with varying zones of (17, 20, 12,17mm) for Gram-positive Staphylococcus aureus) and (10, 22, 10, 22mm) for Gram-negative (E.coli) at (1, 1.5, 1.75, 2mM) respectively. Also, the results showed that green synthetic nanoparticle could heal burns in 12 day compared to 14 days for sliver sulfadiazine and 18 days for negative control. Conclusion: green synthetic nanoparticle possessed antibacterial and anti-inflammatory activity due to active constituents of plant.
