“…Placental TNF could be involved in these processes since hypoxia/re-oxygenation of placental tissues in vitro increased secretion of the cytokine and activation of endothelial cells in a TNF-dependent manner [54]. Although increased apoptosis was also noticed in preeclamptic placentae [152], TNF may not be a primary cause[153]. Stimulation of normal and preeclamptic trophoblasts failed to demonstrate increased programmed cell death but resulted in higher sensitivity of the latter to TNF-induced secretion of tissue factor [154].…”

“…Although no changes in the percentage of Ki67 positive cells and M30 positive cells were observed following short term BPA exposure in this study, previous studies have described anti-proliferative and apoptotic effects of BPA on term primary cytotrophoblast or BeWo cells by measuring [3H]-thymidine incorporation or quantifying changes in apoptotic markers, including M30 and tumor-necrosis factor-alpha protein [ 61 , 72 ]. In this study, we quantified Ki67 protein expression using immunostaining, which is a proliferative marker that is expressed in all active phases of the cell cycle (G1, S, G2, and M); we also used an anti-M30 antibody, which is directed against a specific epitope of cytokeratin 18 that is formed by early caspase cleavage in apoptotic cells [ 97 , 98 ]. Additional markers of apoptosis and proliferation should be explored to further investigate the effects of BPA on trophoblast turnover in cultured explants.…”

“…On the other hand, our results showed that high levels of vitamins C and E, separately or in combination, were able to induce an increase in the production of TNF‐α at the level of protein and mRNA. This major proinflammatory cytokine is in the center of innumerable physiological and pathophysiological processes related to healthy 3,23 and complicated pregnancy 24 . TNF‐α seems to be necessary to fetal development, because it is expressed in normal tissues of various organs during gestation.…”

“…It has been shown to regulate progesterone, estradiol and hCG production in placental fragments, JEG‐3 cells (placental choriocarcinoma) or purified cytotrophoblasts, 25 suggesting its implication in trophoblast differentiation and function. TNF‐α is also involved in apoptosis and turnover of trophoblast, 24,26 and in vascular reactivity by enhancing response to vasoconstrictor molecules such as endothelin 27 or impairing response to vasodilatator molecules such as acetylcholine 28,29 . Therefore, any abnormal production of TNF‐α during pregnancy, in maternal circulation and/or in feto‐placental unit, could lead to adverse pregnancy outcomes such as abortion, intrauterine growth restriction or preeclampsia 30 …”
