Dissociation between force and intracellular sodium activity with strophanthidin in isolated sheep Purkinje fibres.

Abstract: SUMMARY1. We have recorded membrane potential, intracellular Na activity (aika) and force of contraction in sheep Purkinje fibres. Force and aia were recorded continuously and simultaneously during exposure to strophanthidin and its subsequent washing off.2. Exposure to strophanthidin in concentrations of 1-5 x 10-7, 5 x 10-7 and 10-5 M caused an increase in force of contraction which was temporally dissociated from the increase of aba.3. There was hysteresis in the relationship between force and aia when the … Show more

“…The methods employed in this study have been fully described elsewhere (Boyett, Hart & Levi, 1986;Boyett et al 1987). To summarize, Purkinje fibres were dissected from sheep hearts obtained from a local abattoir or from dog hearts obtained at the end of experiments carried out by colleagues.…”

“…In cardiac Purkinje fibres the increase is (in part at least) the result of the rise in aia caused by the block of the Na+-K+ pump by cardiac glycosides (Wasserstrom, Schwartz & Fozzard, 1983;Eisner et al 1984; Vassalle & Lee, 1984;Boyett, Hart & Levi, 1986). However, cardiac glycosides may have another inotropic action (Noble, 1980).…”
