Dissection of T cell antigen receptor signaling using protein tyrosine kinase inhibitors

Abstract: T cell antigen receptor (TcR) recognition of appropriately presented antigen results in the rapid activation of protein tyrosine kinases. Subsequent events include activation of protein kinase C and increased intracellular free calcium which lead to the activation of transcription factors involved in regulating interleukin-2 gene expression. We have assayed the ability of a panel of protein tyrosine kinase (PTK) inhibitors to interfere with activation of the NF-AT transcription factor by TcR ligation or treatm… Show more

“…Finally, the biochemical events leading to a state of unresponsiveness as well as maintaining it are not well characterized; both the calcium influx and PKC activation play a major role (Dubois et al 1994). Stimulation of T cells by PMA and calcium ionophore can at least in part mimic TCR engagement (Baldari & Telford 1994). In this study we attempted to overcome antigen specific unresponsiveness or anergy from several angles.…”

“…Inhibitors of T cell PTKs have been described in the literature which repress T cell activation and T cell-mediated cytotoxicity [83][84][85][86]. However, the fine specificity of these inhibitors are not known.…”
