Disruption of Renal Tubular Mitochondrial Quality Control by Myo-Inositol Oxygenase in Diabetic Kidney Disease

Abstract: Diabetic kidney disease (DKD) is associated with oxidative stress and mitochondrial injury. Myo-inositol oxygenase (MIOX), a tubular-specific enzyme, modulates redox imbalance and apoptosis in tubular cells in diabetes, but these mechanisms remain unclear. We investigated the role of MIOX in perturbation of mitochondrial quality control, including mitochondrial dynamics and autophagy/mitophagy, under highglucose (HG) ambience or a diabetic state. HK-2 or LLC-PK1 cells subjected to HG exhibited an upregulation … Show more

“…However, the manner in which the autophagy process changes in these cells in DN remains unclear. In TECs from diabetic animals, inhibited autophagy was implicated by the investigation of Zhan et al (32), whereas autophagy activation was suggested by the study of Zhao et al (33). In podocytes, Fang et al (34) reported that hyperglycemia induced impaired autophagy, whereas Ma et al (35) demonstrated that high glucose promoted autophagy.…”

“…Previous studies have demonstrated that fragmentation of mitochondria within the kidney is linked to both acute (44) and chronic (37,45,46) renal injury and that it is likely that this is the reason for the inability of the diabetic Aifm1…”

“…A whole-kidney mean glomerular injury score was obtained by averaging scores from all glomeruli on one section. 63 For electron microscopy, kidney cortical tissues were cut into small pieces (1 mm 3 ), fixed with 2.5% glutaraldehyde, and embedded in Epon 812 (Polysciences Inc., Warrington, PA). Transmission electron micrographs were obtained using an EM-10 microscope (Zeiss) operated at 60 kV.…”
