Disruption ofNectin-Like 1Cell Adhesion Molecule Leads to Delayed Axonal Myelination in the CNS

Abstract: Nectin-like 1 (Necl-1) is a neural-specific cell adhesion molecule that is expressed in both the CNS and PNS. Previous in vitro studies suggested that Necl-1 expression is essential for the axon-glial interaction and myelin sheath formation in the PNS. To investigate the in vivo role of Necl-1 in axonal myelination of the developing nervous system, we generated the Necl-1 mutant mice by replacing axons 2-5 with the LacZ reporter gene. Expression studies revealed that Necl-1 is exclusively expressed by neurons … Show more

“…Necls/SynCAMs were shown to be involved in late stages of neural circuit formation, in particular synaptogenesis (Biederer et al, 2002) and myelination in the peripheral (Maurel et al, 2007;Spiegel et al, 2007) and the central nervous system (Park et al, 2008). Here, we demonstrate that Necls/SynCAMs are also involved in early steps of neural circuit formation in the chick spinal cord in vivo.…”

“…Furthermore, some Necl family members were identified as tumor suppressor genes, called Tslc (tumor suppressor in lung cancer) and Tsll (Tslc-like) (reviewed in Takai et al, 2003). More recently, Necl1/SynCAM3 and Necl4/SynCAM4 were shown to be required for myelination in the peripheral and the central nervous system (Maurel et al, 2007;Park et al, 2008;Spiegel et al, 2007). Very little is known about the role of Necl3/SynCAM2.…”

“…G ratio, an indicator of the myelin sheath thickness, is restored to normal levels in Tag-1 Ϫ/Ϫ ;plp mice, suggesting that glial TAG-1 positively regulates the myelination process in the CNS. Therefore, TAG-1 may be added to the list of a small number of proteins (PDGF␣, WAVE-1, NogoA, neuregulin 1 nectin-like 1) that affect the level of myelination in CNS fibers apart from those that influence the myelin structure such as MBP and PLP (Popko et al, 1987;Griffiths et al, 1998;Fruttiger et al, 1999;Kim et al, 2006;Brinkmann et al, 2008;Park et al, 2008;Pernet et al, 2008).…”

“…Axonal SynCAM1 and SynCAM3 interact with SynCAM4 on Schwann cells to promote myelination in the PNS (Maurel et al, 2007;Spiegel et al, 2007). In the CNS, SynCAM2 and SynCAM3 have been implicated in myelination, although their detailed expression in axons versus glia is not clear yet (Kakunaga et al, 2005;Park et al, 2008;Pellissier et al, 2007). However, a role in earlier aspects of neural circuit formation had not been considered.…”
