Dispersed and compact chromatin demonstrated with a new EM method: Phosphotungstic acid hematoxylin block-staining

Abstract: Glutaraldehyde/KMnO4 double fixation and phosphotungstic acid hematoxylin (PTAH) block-staining, before dehydration were found to reveal, with great detail and sharpness, the nuclear distribution of compact heterochromatin masses as electron-lucent patches. By contrast the areas of decondensed and dispersed chromatin acquired a high electron density due to the binding of the large PTAH molecule to basic groups in the loosened chromatin network. The method was tested on human blood leukocytes, on the thymus gla… Show more

“…Speci cally, after deparaf nization and hydration, small pieces of the tissue were re xed in 2.5% glutaraldehyde ( buffered with 0.2 M cacodylate, pH 7.2 ). They were further post xed in 0.25% aqueous potassium permanganate ( KMnO 4 ), treated with 5%oxalic acid, and block-stained for basic proteins with the anionic PTAH reagent, prepared according to Lillie [ 14 ], for 24 h at room temperature, before hydration [ 10,11,15 ].…”

“…It also entails a minimum of manipulations, whereby the in vivo state of the chromatin is well preserved. This is highly desirable when applying the staining procedure of PTAH before dehydration [21].…”

“…Since differential chromatin condensation seems to be the most likely factor determining stainability with PTAH, Issidorides and colleagues [1] explored the possibility of adapting the PTAH method to electron microscopy [1,21]. The implication is that the compact chromatin prevents, through steric hindrance, the penetration and binding of the large molecular weight anionic PTAH reagent (3400 MW) to the histone's basic groups.…”
