Dismutation of dihydrofolate by dihydrofolate reductase

Abstract: Degradation of 7,8-dihydrofolate (H2folate) in the presence of dihydrofolate reductase (DHFR) has been shown due not to an oxygenase activity of the reductase as previously reported but to dismutation of H2folate to folate and 5,6,7,8-tetrahydrofolate (H4folate). The reaction can be followed spectrophotometrically or by analysis of the reaction mixture by high-performance liquid chromatography (HPLC). The products have also been isolated and characterized. Oxygen uptake during the reaction is much less than st… Show more

“…Thymidylate synthetase activity is only expressed in replicating cells and is highest in fastgrowing cells. Consequently, drugs targeted to DHFR, such as methotrexate, have proven to be effective chemotherapeutic agents as they are selective inhibitors of rapidly growing cells [17,18]. Folate coenzymes are also used in two steps of de novo cytosolic purine biosynthesis.…”

“…In the present study we also measured the effect of broccoli consumption on folate level as the latter plays an important role in nucleotide biosynthesis, serine and glycine metabolism, DNA methylation and methionine biosynthesis (Blakley and Cocco 1984). A suboptimal status of folate is often associated with increased risk of cardiovascular disease (Boushey et al 1995) and cancer (Kim 1999).…”

“…MTX is an antifolate and its primary cellular target is dihydrofolate reductase, which catalyzes the reduction of folate and 7,8 dihydrofolate to 5,6,7,8 tetrahydrofolate. 55 An essential requirement for both cellular and viral DNA synthesis is a large expansion in the pools of deoxyribonucleoside triphosphates (dNTPs). 55, 56 It is conceivable that a lowering of intracellular nucleotide pools observed in response to MTX is partially responsible for inhibition of viral replication.…”
