Discovery of Novel Highly Potent Hepatitis C Virus NS5A Inhibitor (AV4025)

Abstract: A series of next in class small-molecule hepatitis C virus (HCV) NS5A inhibitors with picomolar potency containing 2-pyrrolidin-2-yl-5-{4-[4-(2-pyrrolidin-2-yl-1H-imidazol-5-yl)buta-1,3-diynyl]phenyl}-1H-imidazole cores was designed based on the SAR studies available for the reported NS5A inhibitors. Compound 13a (AV4025), with (S,S,S,S)-stereochemistry (EC50 = 3.4 ± 0.2 pM, HCV replicon genotype 1b), was dramatically more active than were the compounds with two (S)- and two (R)-chiral centers. Human serum did… Show more

“…They significantly reduce HCVRNA levels and enhance SVR when given in conjunction with PEG-IFN and RBV. They also result in very high SVR rates among patients with genotype 1 infection when given in combination with other direct-acting antivirals with or without RBV [105] . …”

“…PK studies in mice, rats, and dogs revealed that 88d demonstrated relatively high plasma levels ( C max ), a long half-life, and good oral bioavailability, which was 65% in rats, relative to 11% for 88a . Moreover, 88d was not associated with any significant pathological or clinical observations in rats and dogs during safety, pharmacology, and subchronic toxicity studies …”

“…This is a period often discussed as one of intense migration and nomadism, while the economy is considered to have been based on pastoralism. The Early Bronze Age Yamnaya period spans from 3300 to 2500 cal BCE and is distributed over a vast territory, with regional variation in material culture [26]. The subsistence economy was based primarily on pastoralism-with some scholars suggesting these populations were highly mobile or semi-nomadic [27]-alongside some initial cultivation of wheat, barley, hemp, millet [28].…”
