Disaturated-phosphatidylcholine and Surfactant protein-B turnover in human acute lung injury and in control patients

Abstract: BackgroundPatients with Adult Respiratory Distress Syndrome (ARDS) and Acute Lung Injury (ALI) have low concentrations of disaturated-phosphatidylcholine and surfactant protein-B in bronchoalveolar lavage fluid. No information is available on their turnover.ObjectivesTo analyze disaturated-phosphatidylcholine and surfactant protein-B turnover in patients with ARDS/ALI and in human adults with normal lungs (controls).Methods2H2O as precursor of disaturated-phosphatidylcholine-palmitate and 113C-Leucine as precu… Show more

“…Fractional synthesis rate (FSR), secretion time (ST), and peak time (PT) were calculated as previously reported [21,18]. Briefly, FSR was calculated by dividing the slope of the linear increase of the enrichment of SP-C by the plasma steady-state enrichment of free 13 C-leucine.…”

“…BALF DSPC pools were significantly diminished in I lungs compared with the NI and control lungs. In ARDS patients we recently reported that DSPC amount in tracheal aspirates was markedly reduced and DSPC FSR was 3.1 times higher than in control patients (34), suggesting an overall compensatory DSPC synthetic mechanism that was still not sufficient to replace the DSPC lost during the acute phase of lung injury.…”
