Directed evolution of a glutaryl acylase into an adipyl acylase

Abstract: Semi-synthetic cephalosporin antibiotics belong to the top 10 of most sold drugs, and are produced from 7-aminodesacetoxycephalosporanic acid (7-ADCA). Recently new routes have been developed which allow for the production of adipyl-7-ADCA by a novel fermentation process. To complete the biosynthesis of 7-ADCA a highly active adipyl acylase is needed for deacylation of the adipyl derivative. Such an adipyl acylase can be generated from known glutaryl acylases. The glutaryl acylase of Pseudomonas SY-77 was mutat… Show more

“…Chromosomal DNA was isolated from an overnight culture of P. aeruginosa PAO1 (Holloway collection) as described by Chen and Kuo (4). The open reading frame (ORF) encoding the acylase gene (bases 2638805 to 2636517, obtained from www.pseudomonas.com) (47) was amplified from chromosomal DNA by PCR and cloned into plasmid pMcTNde (45) under the control of the tac promoter, resulting in plasmid pMc-PA2385. For this procedure, the three bases preceding the putative start codon were altered to create an NdeI restriction site.…”

“…A putative acylase gene (the PA2385 gene) was located in the PAO1 genome sequence (47) by homology searching using the sequence encoding the ␤-lactam acylase from Pseudomonas SY-77 (45). This gene is also known as qsc112 (52) and pvdQ (21).…”

“…However, due to precipitation phenomena, it was decided to use the DC protein assay with these purified acylase samples. This resulted in ϳ35% higher protein concentration values and thus in a decreased k cat value for SY-77 WT compared with an earlier study (16).…”

We genetically separated the multiple mutants and measured the AD/GL activity ratio of the resulting mutants in order to pinpoint the mutation that contributes most to the altered activity.…”
