Direct Visualization of Transient Thermal Response of a DNA Origami

Abstract: The DNA origami approach enables the construction of complex objects from DNA strands. A fundamental understanding of the kinetics and thermodynamics of DNA origami assembly is extremely important for building large DNA structures with multifunctionality. Here both experimental and theoretical studies of DNA origami melting were carried out in order to reveal the reversible association/disassociation process. Furthermore, by careful control of the temperature cycling via in situ thermally controlled atomic for… Show more

“…Answering these questions may be important for understanding the mechanism through which 3D DNA box origami can develop resistance to serum. The mica surface may have a stabilizing role for the bottom side of box origami [29]. However, it is worth noting that the damage to box origami in serum solution may be due to the hollow structure.…”

“…AFM evaluates the stability of DNA origami to various stimuli basing its morphologic evolution in situ. [28,29]. However, detailed evidence of the structural stability of the origami, direct visualization of the degradation process, and real-time analyses cannot be achieved with standard AFM because of its slow innate scan speed [30].…”

“…A cooling water fluid circuit refrigerates the piezo-scanner. Before the acquisition of every image, a dwell time of 5 min was allowed to equilibrate the temperature 17 . AFM image acquisition was carried out in tapping mode in air (probes: OMCL-AC160TS-C2, Olympus, Japan).…”

“…Here we use thermally controlled AFM (TC-AFM) 17,18 to directly investigate both the structure and in situ dynamics of water condensation on mica (hydrophilic surface) in both the hydrophobic-hydrophilic interface (graphene and mica) and the hydrophilic-hydrophilic interface (MoS 2 and mica). By carefully monitoring the whole dynamic water growth process, the water interaction within the hydrophobic-hydrophilic interface was investigated.…”

“…Staples bind to the scaffold only, not to other staples, and can hybridize to the same number of scaffold domains at any stage of assembly. Arbona et al [22,26] have presented a model of origami folding in which cooperativity arises from two effects.…”
