Activated layered double hydroxides (LDHs) with high crystallinity, obtained by calcination/rehydration of LDH precursors synthesized by urea decomposition, have higher catalytic activity in acetone self‐condensation and Knoevenagel reactions than less crystalline materials obtained from LDH precursors synthesized by titration co‐precipitation. The activated LDHs possess both basic and acidic sites. High resolution transmission electron microscopy (HRTEM) confirms that the highly crystalline activated LDHs retain the lattice structure of the LDH precursors with lattice parameters a = b = 0.31 ± 0.01 nm and α = 60 ± 2°. An acid‐base catalytic mechanism has been proposed to interpret the catalytic behavior based on the fact that acid‐base hydroxyl group pairs on the activated LDH surface have a separation of 0.31 nm. It is proposed that the active sites are mainly located on the ordered array of hydroxyl sites on the basal surfaces rather than on the edges, as has been previously suggested. © 2007 American Institute of Chemical Engineers AIChE J, 2007

In a prospective phase I/II clinical study, we treated eight patients suffering from recurrent glioblastoma multiform with stereotactically guided intratumoral convection-enhanced delivery of an HSV-1-tk gene-bearing liposomal vector and systemic ganciclovir. Noninvasive identification of target tissue together with assessment of vector-distribution volume and the effects of gene therapy were achieved using magnetic resonance imaging and positron emission tomography. The treatment was tolerated well without major side effects. In two of eight patients, we observed a greater than 50% reduction of tumor volume and in six of eight patients focal treatment effects. Intracerebral infusion of contrast medium before vector application displayed substantial inhomogeneity of tissue staining indicating the need of test infusions to monitor the mechanical distribution of vectors. Visualization of therapeutic effects on tumor metabolism and documentation of gene expression using positron emission tomography indicated that molecular imaging technology appears to be essential for the further development of biological treatment strategies.

In a series of 72 open brain tumor operations the ³²P (radiophosphorus) test proved to be valuable for the removal of visually not detectable tumor residues. As during resection a topographical orientation in the depth of tumor cavities was nearly impossible, a computerized measuring device was developed which enables the surgeon to locate the tumor boundaries by simultaneous comparison with preoperative CT scans. For ³²P-beta emission measurements a miniature semiconductor probe can be attached to the tip of the four-axis digitizing arm allowing the evaluation of radicality by topographic distinction between tumor ramifications and normal brain tissue. The device was tested during 3 craniotomies and the perspectives of computerized stereotaxy are discussed.
