Direct trafficking pathways from the Golgi apparatus to the plasma membrane

Abstract: In eukaryotic cells, protein sorting is a highly regulated mechanism important for many physiological events. After synthesis in the endoplasmic reticulum and trafficking to the Golgi apparatus, proteins sort to many different cellular destinations including the endolysosomal system and the extracellular space. Secreted proteins need to be delivered directly to the cell surface. Sorting of secreted proteins from the Golgi apparatus has been a topic of interest for over thirty years, yet there is still no clear… Show more

“…Moreover, host lipid rafts have been reported to be critically involved in apical targeting, assembly, and virus budding. In this case, the subcellular distribution of lipid raft on internal membranes, including the Golgi apparatus or the ER, has a significant impact in the sorting of proteins and in the trafficking and overall exocytosis of viral proteins, which constitute fundamental steps to support viral infection (Takeda et al, 2003 ; Von Blume and Hausser, 2019 ; Stalder and Gershlick, 2020 ).…”

“…Golgi apparatus is a crucial intracellular organelle that functions as the major protein modification and as a hub for protein transporting and delivering [ 32 , 50 ]. Moreover, the Golgi apparatus is also recognized as an organelle where newly synthesized proteins are post-translationally modified for intracellular trafficking [ 51 , 52 ], facilitating the delivery of polarity proteins toward specific regions of the cell membrane. The biosynthetic pathway by the Golgi is primordial to the secretion of extracellular matrix components, as well as the transmembrane and membrane-bound proteins, to the cell membrane [ 52 , 53 ].…”

The biosynthetic pathway by the Golgi is primordial to the secretion of extracellular matrix components, as well as the transmembrane and membrane-bound proteins, to the cell membrane [ 52 , 53 ]. It has long been identified that dynamic maintenance of cell polarity can be achieved through four distinct approaches: (i) activation gradients of RhoGTPases, (ii) actin cytoskeleton and focal adhesions, (iii) microtubule network and intracellular organelles, and (iv) lipid gradients and cell membrane [ 32 ].…”

“…They collectively participate in the dynamic exchanges necessary for cell homeostasis ( Rothman, 2002 ; Søreng et al, 2018 ). Membrane traffic is tightly coordinated with protein synthesis, signal transduction of environmental stimuli and cytoskeleton organization, allowing the implementation of key cellular functions such as endocytosis, exocytosis, or migration ( McMahon and Gallop, 2005 ; Habtezion et al, 2016 ; Vega-Cabrera and Pardo-López, 2017 ; MacGillavry and Hoogenraad, 2018 ; Margaria et al, 2019 ; Tapia et al, 2019 ; Buratta et al, 2020 ; Stalder and Gershlick, 2020 ).…”
