Direct shoot regeneration from leaf explants of Spilanthes acmella

Abstract: Multiple shoots of Spilanthes acmella Murr. were induced from nodal buds of in vivo and in vitro seedlings on Murashige and Skoog (MS) medium containing 1.0 mg dm -3 6-benzyladenine (BA) and 0.1 mg dm -3 α-naphthaleneacetic acid (NAA). Adventitious shoots were successfully regenerated from the leaf explants derived from the above mentioned multiple shoots. The efficiency of shoot regeneration was tested in the MS medium containing BA, kinetin, or 2-isopentenyl adenine in combination with NAA, indole-3-acetic a… Show more

“…The abaxial epidermal cells increased in size and simultaneously divided into several planes which formed meristemoids on the surface of the midrib. These meristemoids, by further cell divisions gave rise to small protrusions of tissue, which gradually organized into shoot buds similarly as described by Saritha and Naidu (2008) in Spilanthes acmella.…”

“…On the contrary positive synergistic effect of cytokinins like BAP or Kn or TDZ or 2iP or zeatin in combination with an auxin like IAA, or NAA on in vitro shoot regeneration through organogenic differentiation has been reported by several workers (Shen et al 2007, Unda et al 2007, Erisen et al 2010, Ghimire et al 2010, Lin et al 2011, Wadl et al 2011, Mandal and Laxminarayana 2012, Perez-Jimenez et al 2012and Bhagya et al 2013. Superior effect of BAP for shoot multiplication has been reported among various medicinal plants of Asteraceae, like Tanacetum cinerariifolium (Hedayat et al 2009), Spilanthes acmella (Saritha and Naidu 2008), Launaea sarmentosa (Mahesh et al 2012). As in the present study, the reduction of shoot regeneration potential by the combined effect of BAP and GA 3 compared to a medium with cytokinin alone was also demonstrated in Guizotia abyssinica (Baghel and Bansal 2014).…”

“…In this work, the leaf explants produced buds mainly from the petiole base. Saritha and Naidu (2008) cultured Spilanthes acmella on MS medium supplemented with different plant growth regulators for direct regeneration from mature leaves or leaves of in vitro grown plantlets. The formation of shoots was observed after 45 d either in the midrib region or basal petiole region or apical region of the leaf.…”
