Direct Shoot Regeneration from Callus of &amp;lt;i&amp;gt;Melicope lunu-ankenda&amp;lt;/i&amp;gt;

Phenolic acids and flavonoids, important bioactive compounds of polyphenols, play a significant role in plants; their impact, mainly as antioxidants, on human health have been of great interest in recent years. The genetically uniform shoots of Eryngium alpinum L. cultured in vitro, developed via axillary buds and regenerated from callus tissue, maintained on the media supplemented with various plant growth regulators, were subjected to the phenolic acids and flavonoids quantitative analysis applying HPLC-DAD technique. In vitro cultures give the opportunity to perform the phytochemical studies on the protected species without harvesting the plant material from natural environment. Depending on the hormonal supplementation, the biomass from the shoot cultures accumulated from 11.41 to 25 times more phenolic acids [the total content ranged from 158.66 to 1817.96 mg/100 g of dry weight (DW)] and from 6.8 to 11.8 times more flavonoids (the total content ranged from 29.30 to 51.30 mg/100 g DW) than the shoots from the soil-grown plant. The polyphenols present in the shoot cultures include two phenolic acids: 3,4-dihydroxyphenylacetic and caffeic, four depsides: caftaric, neochlorogenic, chlorogenic, isochlorogenic, and rosmarinic acids, and flavonoids: aglycone-isoquercetin and glucoside-quercitrin. Most of them (apart from chlorogenic and rosmarinic acids) were detected for the first time in this species cultured in vitro. To our best knowledge, the present report is the first one that discusses establishment of Eryngium alpinum L. in vitro cultures and the shoot and callus biomass capacity to produce two subgroups of polyphenols i.e. phenolic acids and flavonoids. Key message This is the first report on establishment of Eryngium alpinum in vitro cultures and the shoot and callus biomass capacity to produce polyphenols-phenolic acids and flavonoids.

The African Sandalwood plant (Osyris lanceolata ) is a threatened shrub or a small hemi-parasitic tree endemic to East Africa and South African regions, which is being severely affected by uprooting for oil extraction, poor natural regeneration, phenological structures (dioecious), medicinal values, lack of sexual recruitment, habitat loss, anthropogenic and climate factors. It has been found that through application of in situ conservation of natural trees with respect to rapid human population growth, the available natural strands of valuable plants such as African sandalwood have not been able to meet the demands of the people in world specifically developing countries. However, advances in plant biotechnology provide new options for collection, multiplication and short-to long-term conservation of Osyris lanceolata species, using in vitro culture techniques. Different aspects of biotechnological applications can be extensively used to reduce the risk of extinction of this valuable plant species and to improve the quality and quantity of essential oils produced by it. Therefore, tissue culture appears to be a promising approach for the propagation and conservation of African sandalwood plant.

The conservation of threatened exceptional plants, which cannot be conserved by seed banking, requires in vitro technologies for many of the approaches needed for their long-term ex situ conservation. This study evaluated the current in vitro plant literature, as represented in Web of Science, to determine its taxonomic overlap with the families and genera of the 775 species currently listed as exceptional. Web of Science was searched using the terms micropropagation, somatic embryogenesis, zygotic embryo, and cryopreservation, and the target genera and families were identified in the more than 19,000 articles evaluated. There were five families with significant overlap between the in vitro literature and exceptional species: Fabaceae, Asteraceae, Orchidaceae, Arecaceae, and Rutaceae. However, there was less overlap at the level of genus, with Citrus, Coffea, and Quercus having the most articles. Significant gaps were also found, with 14 exceptional families and half of the exceptional genera having no representation in the Web of Science search results. The 20 exceptional species with the most articles were all economically important species, and these had 343 threatened congeners that could be prioritized for research. A highly important group of exceptional plants that was significantly under-represented in the literature was tropical woody species, which form the backbone of the diversity of the world’s threatened rainforests. Overall, there are areas of strength upon which to build future work, but significant gaps where research should be prioritized for effectively conserving exceptional plants.
