Direct Recognition of Cytomegalovirus by Activating and Inhibitory NK Cell Receptors

Abstract: Natural killer (NK) cells express inhibitory receptors for major histocompatibility complex (MHC) class I antigens, preventing attack against healthy cells. Mouse cytomegalovirus (MCMV) encodes an MHC-like protein (m157) that binds to an inhibitory NK cell receptor in certain MCMV-susceptible mice. In MCMV-resistant mice, this viral protein engages a related activating receptor (Ly49H) and confers host protection. These activating and inhibitory receptors are highly homologous, suggesting the possibility that … Show more

“…8). This model provides an explanation for the skewed repertoire in adaptive NK cells seen in HCMV + individuals towards inhibitory KIR that recognize self HLA class I. Activating KIRs could play a similar role to NKG2C and recognize unknown HCMV‐induced ligands, similarly to Ly49H/m157 in mouse 129, 130…”

“…Certain KIRs and Ly49 proteins recognize MHC class I molecules. Other NK-cell receptors recognize a broad range of stress-induced molecules and other non-classical MHC molecules 63,64 , as well as viral gene products 65,66 and other NK-cell receptors 67 . NK-cell activity has a broad phylogenetic distribution extending to invertebrates 68 .…”

“…Inhibitory receptors of mouse NK cells comprise several Ly49 receptors, CD94/NKG2A [7] or CD48 [8]. Activating receptors such as Ly49D [9], Ly49H [10] or NKp46 [11] recognize nonself molecules that are expressed upon infection. Another type of an activating surface molecule is natural killer group 2D (NKG2D).…”
