Direct dorsal hippocampal–prelimbic cortex connections strengthen fear memories

Abstract: The ability to regulate the consolidation and strengthening of memories for threatening experiences is critical for mental health, and its dysregulation may lead to psychopathologies. Re-exposure to the context in which the threat was experienced can either increase or decrease fear response through distinct processes known, respectively, as reconsolidation or extinction. Using a context retrieval-dependent memory enhancement paradigm in rats, we report that memory strengthens through the activation of direct … Show more

“…In addition to the ACC to vHPC circuit we identified, a neural circuit composed of the mPFC, the thalamic NR, and the hippocampus has been demonstrated to mediate memory generalization (Xu and Südhof, 2013). It is known that there are direct projections from vHPC to mPFC (Ye et al, 2017;Marek et al, 2018); thus, there may be a close relationship between the ACC to vHPC circuit and the mPFC-NR-hippocampus circuit, which needs to be investigated in the future. “…Mice were briefly headrestrained while the stainless-steel obturator was removed and a microinjection tube (30 gauge, 4.0 mm, RWD Life Science) was inserted into the guide cannula. CNO (0.5 mM; Ye et al, 2017) was infused into the vHPC through the microinjection tube at a flow rate of 0.15 l/min and the injection volume was 0.8 l per site.…”

“…Declarative memories are initially stored within the dHP (recent memory) and, over time, slowly transferred to the mPFC for perma-nent storage (remote memory) via the EC (18)(19)(20). It has been reported that a small population of dCA1 neurons directly project to the PL (9)(10)(11). However, the role of these monosynaptically targeted PL neurons in remote memory is not clear, given the technical difficulty in manipulating functionally specific PL neurons (PL engram cells) receiving direct dCA1 projection.…”

“…Abundant literature links theta frequency coupling within prefrontal-hippocampal networks to cognitive performance and emotional states of adults (Adhikari et al, 2010; Xu and Sudhof, 2013; Spellman et al, 2015; Hallock et al, 2016; Place et al, 2016; Ye et al, 2017). The early emergence of directed communication between PFC and i/vHP raises the question of functional relevance of this early coupling during development and at adulthood.…”
