Direct detection of cancer biomarkers in blood using a “place n play” modular polydimethylsiloxane pump

Abstract: Cancer biomarkers have significant potential as reliable tools for the early detection of the disease and for monitoring its recurrence. However, most current methods for biomarker detection have technical difficulties (such as sample preparation and specific detector requirements) which limit their application in point of care diagnostics. We developed an extremely simple, power-free microfluidic system for direct detection of cancer biomarkers in microliter volumes of whole blood. CEA and CYFRA21-1 were chos… Show more

“…PDMS slab at the outlet pulls the liquid further into the channel via degassing mechanism. 62 The sedimentation area is a trench like structure which is deeper and wider than other areas to slow down the flow rate and allows blood cells to sediment under gravity. All the reagents including plasma, wash buffers, and detection antibody-gold-silver complex were pumped to the reaction site at different times with the PDMS pump.…”

“…Microfluidic technology has broad applications in basic research and clinical analysis due to its portability, affordability, and high sensitivity (Nora Dickson et al, 2011;Zhang et al, 2013;Zhao et al, 2013). It holds great promise as it can perform typical laboratory operations using a small volume of reagents in less time (Teh et al, 2008).…”

“…Passive separation techniques can be stored under pressure or driven by capillary forces [ 13 , 14 , 15 ]. Using capillary forces in plasma separation is more desirable, since it demands neither vacuum packaging, nor degassing of a suction material [ 16 ]. The mechanism of separation is generally based on sedimentation, size exception, or a combination of them [ 17 ].…”
