Dipyridamole potentiates the in vitro activity of MTA (LY231514) by inhibition of thymidine transport

Abstract: The novel pyrrolopyrimidine-based antifolate LY231514 (MTA), inhibits multiple folate-requiring enzymes including thymidylate synthase, glycinamide ribonucleotide formyltransferase and dihydrofolate reductase. Both thymidine and hypoxanthine are required to reverse MTA growth inhibition in leukaemia and colon cancer cells. Prevention of MTA growth inhibition by thymidine and/or hypoxanthine was investigated in two human lung (A549, COR L23) and two breast (MCF7, T47D) tumour cell lines, and the effect of the n… Show more

“…In this study, both cell lines were relatively resistant to MTA in comparison with 211H cells with the IC 50 values for A549 cells being consistent with previous studies [42,43] . To elucidate mechanisms of resistance to MTA, we performed quantitative RT-PCR analysis using TS , FPGS and RFC1 genes.…”

“…Several descriptions of the kinetics and pharmacological properties of nucleoside transporters in the cell have been published (19, 29 -31). Multiple drugs can inhibit the function of these carriers (32,33). Decreasing the rate of renal reabsorption of thymidine by chemically inhibiting nucleoside carriers could lead to a significant decrease in thymidine concentrations in blood of MNGIE patients.…”

“…We used a modified rapid mixing technique, combined with an inhibitor-stop method, to determine thymidine uptake into L1210 murine leukemia cells, as described previously (14,17 3 H]thymidine in the extracellular space that was carried through the oil layer.…”
