Diphtheria toxin endocytosis and membrane translocation are dependent on the intact membrane-anchored receptor (HB-EGF precursor): studies on the cell-associated receptor cleaved by a metalloprotease in phorbol-ester-treated cells

Abstract: Preincubation of Vero cells with 1 microM phorbol 12-myristate 13-acetate (PMA) decreased the specific binding of diphtheria toxin by about 50%, whereas the toxic effect, endocytic uptake and membrane translocation were completely blocked. Toxin bound to PMA-treated cells was released upon incubation with heparinase. The effect of PMA was abrogated in the presence of EDTA or N-(DL-[2-(hydroxyaminocarbonyl)methyl]-4-methyl-pentanoyl)-L-3-(2' - naphthyl)-alanyl-L-alanine 2-aminoethyl-amide (TAPI), a specific inh… Show more

“…MMPs are reported to promote cell proliferation by releasing local growth factors stored within the extracellular matrix and process growth factor receptors. 34,35,46 In the present study, however, MMP-2 derived from CD31(Ϫ) CD45(Ϫ) SP cells did not stimulate the proliferation of myoblasts in vivo ( Figure 7D). The factors that stimulate the proliferation of myoblasts remain to be determined in a future study.…”

“…[33][34][35][36] Metalloproteases (MMP) are implicated in the cleavage of proHB-EGF and metalloprotease inhibitors (TIMP) prevent this process. 37 The presence of MMP and TIMP in pterygium is well documented. 38 -42 The interaction between HB-EGF, MMP, and TIMP in pterygia is likely to result in the liberation of the soluble form of HB-EGF from pterygium epithelium.…”

“…4B). As an inhibitor control we used diphtheria toxin, which is taken up by clathrin-mediated endocytosis (13). Diphtheria toxin-induced cell rounding of HeLa cells was blocked in the presence of chlorpromazine (Fig.…”
