Diphtheria Toxin-Antitoxin and Toxoid

Abstract: N. Y. wIESE two preparations, toxin-antitoxin and toxoid, are now being widely used for immunization of infants and young children against diphtheria. Toxin-antitoxin has been longest in use. Behring was the first to employ it experimentally as an immunizing injection in man. His preparation was the undiluted toxic broth with its toxin nearly neutralized by antitoxin. He never gave a clear explanation of how he prepared and standardized it. The breaking out of the war delayed the practical utilization of toxin… Show more

“…Jedinjenja aluminijuma kao adjuvansi u humanim vakcinama se koriste od 1932. godine (27). Prvi upotrebljen adjuvans na bazi aluminijuma bio je aluminijum kalijum sulfat, koji je zbog slabe reproducibilnosti u potpunosti zamenjen aluminijum hidroksidom i aluminijum fosfatom, koji se mogu pripremiti standardizovanim metodama i za koje se antigen vezuje direktnom adsorpcijom (28).…”

“… 1927: Fractional purification of toxoid by precipitation with aluminum salts provided robust immunization and protection in guinea pig [ 37 ]. 1932: Alum greatly enhanced the value of diphtheria toxoid immunization in humans [ 38 ]. 1933: A single dose of alum-precipitated toxoid afforded protective immunity in humans, assessed by the Director of the National Institutes of Health (NIH) as being much more effective and tolerable compared to toxoid alone [ 39 ].…”

“…All patients had received at least 1 acellular pertussis vaccination before study entry. However, baseline values were certainly influenced by maternal IgG 25,26 because the mean age of patients at the time of enrollment was 2.9 months. To best assess response to a series of immunizations while receiving corticosteroid therapy, antibody titers to tetanus and diphtheria were measured 3 months after completion of prednisolone therapy.…”
